摘  要:丝裂原活化蛋白激酶MAPK(mitogen-activated protein kinase)通路对细胞增殖、迁移、分化和凋亡等具有重要的调控作用,它们也参与调节细胞基因表达和代谢。依据亚基结构和调控特征,将哺乳动物细胞MAPKs分为"经典"和"非典型"。ERK3(extracellular signal-regulated kinase 3)也称为MAPK6,属于非典型MAPKs,仅具有一个磷酸受体位点。新近研究提示,ERK3在肿瘤的发生发展中发挥重要作用,可能成为肿瘤治疗的新靶点。该文对ERK3的转录后及翻译后修饰、生物化学和生物学功能,及其在肿瘤中的表达及作用进行综述,为寻找肿瘤治疗新策略提供思路。Mitogen-activated protein kinase(MAPK)pathway plays an important role in the regulation of cell proliferation,migration,differentiation and apoptosis.They are also involved in regulating gene expression and metabolism in cells.According to the subunit structure and regulatory characteristics,mammalian MAPK is classified into classical and atypical.ERK3(also known as MAPK6)belongs to the atypical MAPK group.ERK3 is characterized by a unique phosphoric acid receptor site.Little is known about the biochemical and biological functions of ERK3.Recent studies suggested that ERK3 plays an important role in the development and progression of tumors,and ERK3 may be a new target for tumor therapy.This review discussed post-transcriptional and post-translational modifications of ERK3.We summarized the biochemical and biological functions of ERK3.At the same time,we reviewed the expression and role of ERK3 in tumors.Elucidating the regulation and function of ERK3 will provide new strategies for tumor therapy.
