摘  要:从天然生物絮凝剂资源菲律宾蛤仔黏附污泥中分离获得1株新的高絮凝活性菌株GHS18,絮凝率为75.1%。综合序列比对和生理生化特征结果,GHS18被鉴定为Pseudoalteromonas paragorgicola。通过Illumina测序平台对菌株GHS18进行全基因组框架扫描测序,基于基因组的分子水平来探索菌株GHS18产絮凝活性物质的基础。生物信息学分析结果表明菌株主要参与淀粉、蔗糖、果糖、甘露糖、半乳糖、氨基糖、核苷酸糖、肽聚糖、脂多糖的生物合成与代谢,具有絮凝多糖合成途径中重要的前体底物(UDP-Glucose、GDP-mannose和UDP-glucuronate)和重复寡糖单元(UDP-Gal、dNTP-D-Glu、dNTP-L-Rham、UDP-3-O-(3-acetyl)-GlcNAc),将重复寡糖单元聚合成多糖的eps基因簇并未发现,推测其具有类似的基因簇代替行使功能。本研究为未来开发该菌株为发酵产品提供了一定的理论依据,也为更进一步的研究奠定了基础。A new bioflocculant-producing strain GHS18 with 75.1%of flocculation rate was isolated from Ruditapes philippinarum conglutination mud(RPM)which as natural bioflocculant resource.The strain GHS18 was identified as Pseudoalteromonas paragorgicola according to the results of sequence blast and biochemical tests.In order to study the bioflocculant-producing basis at genomic level,the scaffold sequences of complete genome were sequenced by Illumina sequencing platform.The bioinformatic analysis indicated that the genome of P.paragorgicola GHS18 was related to starch,sucrose,fructose,mannose and galactose uptake system,and metabolism of peptidoglycan,lipopolysaccharide,amino sugar and nucleotide sugar.The key precursors(including UDP-Glucose,GDP-mannose and UDP-glucuronate),oligosaccharide repeating units(including UDP-Gal,dNTP-D-Glu,dNTP-L-Rham and UDP-3-O-(3-acetyl)-GlcNAc)in the biosynthesis of extracellular polysaccharide were found.However,the eps gene cluster which polymerized repeating units was not found,it is speculated that it has a similar gene cluster instead of functioning.This study provides a theoretical basis for the future development of this strain for fermentation products,and also laid the foundation for further research.
