摘 要:目的探究有丝分裂激酶A(Aurora-A)调控间充质干细胞向肝癌相关成纤维细胞转化对肝癌细胞增殖、侵袭和迁移的影响。方法将HepG2及hAD-MSCs细胞分为HepG2组、hAD-MSCs组、hAD-MSCs+HepG2组、NC组和Aurora-A-shRNA组。Western blot法检测hAD-MSCs组、hAD-MSCs+HepG2组、NC组和Aurora-A-shRNA组hAD-MSCs细胞中肿瘤相关性成纤维细胞(CAF)相关蛋白α平滑肌肌动蛋白(α-SMA)、肌间线蛋白(Desmin)、成纤维细胞激活蛋白(FAP)、血小板反应蛋白/凝血酶敏感蛋白1(Tsp-1)、成纤维细胞特异性蛋白(FSP)表达水平。CCK-8法、Transwell法和Western blot法分别检测HepG2组、hAD-MSCs+HepG2组、NC组和Aurora-A-shRNA组HepG2细胞增殖抑制率、侵袭和迁移能力、Aurora-A表达。结果与hAD-MSCs组相比,hAD-MSCs+HepG2组和NC组hAD-MSCs细胞中CAF相关蛋白α-SMA、Desmin、FAP、Tsp-1、FSP表达水平显著增加(P<0.05)。与hAD-MSCs+HepG2组相比,Aurora-A-shRNA组α-SMA、Desmin、FAP、Tsp-1、FSP表达水平显著降低(P<0.05)。与HepG2组相比,hAD-MSCs+HepG2组和NC组HepG2细胞内侵袭和迁移细胞数显著增加,增殖抑制率显著降低(均P<0.05)。与hAD-MSCs+HepG2组和NC组相比,Aurora-A-shRNA组HepG2细胞中Aurora-A表达水平、侵袭和迁移细胞数显著降低,增殖抑制率显著升高(均P<0.05)。结论在肿瘤微环境中,Aurora-A低表达对肝癌细胞增殖、侵袭和迁移的抑制作用与其抑制hAD-MSCs向CAFs转化有关。Objective To explore the effect of Aurora-A on the proliferation,invasion and migration of hepatocellular carcinoma cells by regulating the transformation of mesenchymal stem cells into hepatocellular carcinoma associated fibroblasts.Methods The experiment was divided into HepG2 group,hAD-MSCs group,hAD-MSCs+HepG2 group,NC group and Aurora-A-shRNA group.Western blot was used to detect the expression of carcinoma-associated fibroblast(CAF)-related proteinsα-SMA,Desmin,FAP,Tsp-1 and FSP in hAD-MSCs group,hAD-MSCs+HepG2 group,NC group and Aurora-A-shRNA group.CCK-8 assay,Transwell method and Western blot were used to detect the proliferation inhibition rate,the invasion and migration abilities,and the expression of Aurora-A in HepG2 cells in HepG2 group,hAD-MSCs+HepG2 group,NC group and Aurora-A-shRNA group,respectively.Results Compared with hAD-MSCs group,the expression levels of CAF-related proteinsα-SMA,Desmin,FAP,Tsp-1 and FSP were significantly increased in hAD-MSCs+HepG2 group and NC group(P<0.05).Compared with hAD-MSCs+HepG2 group,the expression levels ofα-SMA,Desmin,FAP,Tsp-1,and FSP were significantly decreased in Aurora-A-shRNA group(P<0.05).Compared with HepG2 group,the numbers of invasion and migration cells were significantly increased in hAD-MSCs+HepG2 group and NC group,and the proliferation inhibition rate was significantly decreased(P<0.05).Compared with hAD-MSCs+HepG2 group and NC group,the expression level of Aurora-A,the numbers of invasion and migration cells were significantly decreased in Aurora-A-shRNA group,while the proliferation inhibition rate was significantly increased(P<0.05).Conclusion In the tumor microenvironment,the inhibitory effect of Aurora-A low expression on the proliferation,invasion and migration of liver cancer cells is related to its inhibition of transformation of hAD-MSCs into CAFs.
