摘  要:目的探讨不同浓度七氟醚预处理对小鼠心肌缺血/再灌注(myocardial ischemia/reperfusion,MI/R)损伤后血浆脂联素(APN)水平的影响。方法将60只C57野生型小鼠随机分为5组:空白对照组、1%七氟醚预处理组、2%七氟醚预处理组、3%七氟醚预处理组、4%七氟醚预处理组。使用不同浓度七氟醚处理各组小鼠后,建立MI/R模型。分别于术后0,6,12,24 h使用ELISA试剂盒测定小鼠血浆脂联素浓度。结果与空白对照组相比,1%七氟醚预处理组、2%七氟醚预处理组、3%七氟醚预处理组血浆脂联素水平显著增加,而4%七氟醚预处理组血浆脂联素水平显著降低(均P<0.05)。在24 h内,随着再灌注的时间增加,血浆脂联素水平逐步降低。结论低浓度七氟醚预处理(1%-3%)可增加小鼠心肌缺血/再灌注损伤后的血浆脂联素浓度,而高浓度七氟醚预处理(4%)可降低其血浆脂联素水平。Objective To investigate the effects of sevoflurane preconditioning with different concentrations on plasma adiponectin level in myocardial ischemia/reperfusion injury mice.Methods Sixty C57 mice were randomly divided into five groups:control group,1%sevoflurane preconditioning group,2%sevoflurane preconditioning group,3%sevoflurane preconditioning group,and 4%sevoflurane preconditioning group.After treatment with different concentrations of sevoflurane,the MI/R model was established.ELISA kit was used to determine the plasma adiponectin level in mice at 0,6,12,24 h after operation.Results Compared with blank control group,the plasma adiponectin level was significantly increased in 1%sevoflurane preconditioning group,2%sevoflurane preconditioning group and 3%sevoflurane preconditioning group,while the plasma adiponectin level was significantly decreased in 4%sevoflurane preconditioning group(all P<0.05).Within 24 h,the plasma adiponectin level was gradually decreased as the time of reperfusion increased.Conclusion Low-concentration sevoflurane(1%-3%)preconditioning can increase the plasma adiponectin concentration after myocardial ischemia/reperfusion injury in mice.However,high-concentration sevoflurane(4%)preconditioning can decrease the plasma adiponectin level.
