摘  要:目的探讨大黄素对糖尿病性白内障大鼠晶状体氧化应激和炎症水平的影响及分子机制。方法将SD大鼠随机分为对照组、链脲佐菌素(streptozotocin,STZ)组、大黄素低(30mg/kg)、中(60mg/kg)和高(90mg/kg)剂量组。后4组大鼠经腹腔一次性注射STZ(65mg/kg)建立糖尿病大鼠模型,血糖值>16.7mol/L为造模成功。造模成功后,立即给予大黄素低、中、高剂量组大鼠大黄素灌胃治疗,对照组与模型组给予等量0.9%氯化钠溶液,每天1次,连续8周。每周对大鼠进行体质量和血糖测量,裂隙灯显微镜观察白内障阶段(0~4)。8周结束后,收集大鼠双侧眼球晶状体,ELISA测定晶状体中超氧化物歧化酶(superoxide dismutase,SOD)活性、丙二醛(malondialdehyde,MDA)浓度、白细胞介素-1(IL-1β)和肿瘤坏死因子(tumor necrosis factor,TNF-α)水平,Western blot法检测晶状体内Keap1和Nrf2蛋白表达。结果与对照组比较,STZ组大鼠体质量降低,血糖升高,晶状体浑浊,ELISA显示氧化应激与炎症水平升高,Western blot法显示晶状体中Keap1蛋白水平升高,Nrf2蛋白水平降低;而与STZ组比较,大黄素治疗组大鼠体质量增加,血糖降低,晶状体浑浊减轻,氧化应激与炎症水平降低,Keap1表达下调,而Nrf2表达上调。结论大黄素可缩短糖尿病性白内障大鼠晶状体混浊时间同时减慢其病程进展,并有一定的浓度依赖性,这可能是通过调控Keap1-Nrf2通路减轻了糖尿病性白内障大鼠晶状体的氧化应激和炎症水平来实现的。Objective To explore the effect of emodin on the level of oxidative stress and inflammation in the lens of diabetic cataract rats and its molecular mechanism.Methods SD rats were randomly divided into control group,streptozotocin(STZ)group,emodin low(30mg/kg),medium(60mg/kg)and(90mg/kg)high-dose groups.In the last 4 groups of rats,a STZ(65mg/kg)was established in the abdominal cavity of the rats to establish a diabetic rat model,and the blood glucose value>16.7mol/L was considered successful.After the model was successfully established,rats in the low,medium,and high dose emodin groups were given intragastric administration immediately.The control group and the model group were given the same amount of normal saline once a day for 8 weeks.The weight and blood glucose of the rats were measured every week,and the cataract stage(0-4)was observed with a slit lamp microscope.After 8 weeks,the lens of both eyes of the rat was collected,and the superoxide dismutase(SOD)activity,malondialdehyde(MDA)concentration,interleukin-1β(IL-1β)in the lens were measured by ELISA.And tumor necrosis factor(TNF-α)levels,Western blot detection of Keap1 and Nrf2 protein expression in the lens.Results Compared with the control group,rats in the STZ group decreased body weight,increased blood sugar,and lens opacity.ELISA showed increased levels of oxidative stress and inflammation.Western blot showed increased levels of Keap1 protein and decreased levels of Nrf2 protein in the lens.Compared with the STZ group,rats in the emodin treatment group gained weight,lowered blood sugar,reduced lens opacity,and reduced oxidative stress and inflammation.The expression of Keap1 was down-regulated,while the expression of Nrf2 was up-regulated.Conclusion Emodin can shorten the lens opacity time of diabetic cataract rats and slow down the course of the disease,and has a certain concentration-dependent.This may be through regulating the Keap1-Nrf2 pathway to reduce the oxidative stress and the lens of diabetic cataract rats.
