高脂饮食致ApoE^(-/-)小鼠动脉粥样硬化及主动脉瓣钙化的实验研究

摘  要:目的探讨高脂饮食致ApoE^(-/-)小鼠动脉粥样硬化及主动脉瓣钙化模型的病理特点。方法10只8周龄C57BL/6J小鼠纳入正常组,20只8周龄ApoE^(-/-)小鼠随机分为对照组和模型组。正常组和对照组给予普通饮食,模型组给予高脂饮食,连续喂养24周。各组小鼠行超声心动图检测。实验结束处死小鼠后检测血清中总胆固醇(total cholesterol,TC)、总甘油三酯(total triglyceride,TG)、低密度脂蛋白胆固醇(low-density lipoprotein cholesterol,LDL-C)、高密度脂蛋白胆固醇(high-density lipoprotein cholesterol,HDL-C)及氧化型低密度脂蛋白(oxidized low-density lipoprotein,oxLDL)水平。油红O染色法检测小鼠主动脉斑块及肝脏脂质蓄积。免疫组织化学法检测小鼠主动脉瓣oxLDL的表达。结果与对照组相比,模型组小鼠血清中TC、TG、LDL-C及oxLDL水平显著升高,HDL-C水平降低;主动脉瓣跨瓣血流峰速升高;主动脉粥样硬化斑块增多;肝脏脂质沉积增加;主动脉瓣oxLDL的表达增加。结论高脂饮食致ApoE^(-/-)小鼠动脉粥样硬化模型伴有明显主动脉瓣钙化,且oxLDL在主动脉瓣表达显著增加。Objective To explore the pathological characteristics of ApoE^(-/-)mice atherosclerosis and aortic valve calcification model induced by high-fat diet.Methods Ten 8-week-old C57 BL/6 J mice were taken as the normal group,and 208-week-old ApoE^(-/-)mice were randomly divided into control group and model group.The normal group and control group were given ordinary diet,and the model group was given high-fat diet for 24 weeks.Each group of mice was tested by echocardiography.After the experiment,the mice were sacrificed,and serum total cholesterol(TC),total triglyceride(TG),low-density lipoprotein cholesterol(LDL-C),high-density lipoprotein cholesterol(HDL-C)and oxidized low-density lipoprotein(oxLDL)levels were tested.Oil red O staining was used to detect aortic plaque and liver lipid accumulation.Immunohistochemistry was applied to detect the expression of oxLDL on mice aortic valve.Results Compared with the control group,the serum levels of TC,TG,LDL-C and oxLDL in the model group increased significantly,while the HDL-C level decreased.The peak velocity of the aortic valve transvalvular blood flow increased.Increased aortic atherosclerotic plaque;Increased liver lipid deposition;Increased aortic valve oxLDL expression.Conclusion The high-fat diet induced ApoE^(-/-)mice atherosclerosis model is accompanied by obvious aortic valve calcification,and the expression of oxLDL in the aortic valve is significantly increased.
