摘 要:目的:研究黄芪甲苷与阿魏酸不同比例组合物对ApoE-/-小鼠动脉粥样硬化(AS)的影响,中药单体给药剂量与在体药效之间的相关性。方法:50只ApoE-/-小鼠随机分为模型组、黄芪甲苷-阿魏酸(HA)(1∶1、2∶1、3∶1、5∶1)组,每组各10只,同时选用10只C57BL/6J小鼠作为正常组。高脂饮食喂养诱导ApoE-/-小鼠建立AS模型;试剂盒测定血清、肝脏TC、TG、LDL-c、HDL-c、ET-1、NO、FFA、FC含量;油红O染色检测肝脏、主动脉、主动脉窦脂质沉积;HE染色观察肝脏、主动脉病理改变;Image J软件统计主动脉窦脂质沉积、主动脉厚度、肝脏油红脂滴及肝脏脂滴空洞的面积。结果:高脂喂养12周后,与正常组比较,模型组血清TC、TG、LDL-c、HDL-c浓度显著升高(P<0.01);与模型组比较,HA不同比例组血清TC、TG、LDL-c含量显著降低(P<0.05)。高脂喂养16周后,与正常组比较,模型组血清、肝脏TC、TG、LDL-c、HDL-c、ET-1、NO、FFA、F C浓度显著升高(P<0.01);肝脏、主动脉、主动脉窦脂质显著沉积(P<0.01);主动脉异常增生并伴有大量胆固醇结晶和泡沫细胞浸润。与模型组比较,HA不同比例组血清TC、TG、LDL-c、ET-1、NO含量显著降低(P<0.05,P<0.01);肝脏TC、TG、FFA、FC浓度显著降低(P<0.01);肝脏、主动脉、主动脉窦病变减轻。结论:HA不同比例组合物AS小鼠都有一定的治疗作用,其中HA 5∶1组效果最显著。Objective:To study the effects of AstragalosideⅣand Ferulic acid composition in different proportion on atherosclerosis(AS)in ApoE-/-mice and the correlation between the dosage of traditional Chinese medicine and its efficacy.Methods:Fifty ApoE-/-mice were randomly divided into model group,AstragalosideⅣand Ferulic acid(HA)(1∶1,2∶1,3∶1,5∶1)groups,each with 10 mice,at the same time,10 C57BL/6J mice were selected as the normal group;ApoE-/-mice were fed a high-fat diet to establish AS model.Kit was used to detect the levels of TC,TG,LDL-c,HDL-c,ET-1,NO,FFA,FC in serum and liver;Oil red O staining was used to detect lipid deposits in the liver,aortic sinus,and the whole aorta;HE staining was used to observe the pathological changes of liver and aorta;Image J software was used to calculate aortic sinus lipid deposition,aortic wall thickness,liver oil red lipid droplets and the area of the cavities of liver lipid droplets.Results:After 12 weeks of high-fat fed,compared with the normal group,the serum TC,TG,LDL-c,HDL-c levels in the model group were significantly increased(P<0.01);Compared with the model group,the serum TC,TG,LDL-c in the HA different ratio group were reduced significantly(P<0.05).After 16 weeks of high-fat fed,compared with the normal group,the serum and liver levels of TC,TG,LDL-c,HDL-c,ET-1,NO,FFA,and FC in the model group increased significantly(P<0.01);Aortic,liver and aortic sinus lipids plasma deposition obviously(P<0.01);The aortic wall was abnormally thickened,accompanied by a large number of cholesterol crystals and foam cell infiltration.Compared with the model group,the level of TC,TG,LDL-c,ET-1,NO in serum on HA groups sigificantly reduced(P<0.05,P<0.01).The concentration of TC,TG,FFA,and FC in liver was reduced(P<0.01).The pathological changes of liver,aorta,and aortic sinus were relatively reduced.Conclusion:HA in different proportion composition have a certain protective effect on AS mice and HA 5∶1 group has the most significant effect.
