摘 要:目的探讨miR-141介导NF-κB信号通路对卵巢癌细胞SKOV3对紫杉醇敏感性的影响,并探讨其作用机制。方法在卵巢癌细胞SKOV3中转染miR-141抑制物设为anti-miR-141组,以转染无关序列的SKOV3细胞设为NC组,以未转染的SKOV3细胞设为Blank组,采用实时荧光定量PCR(qRT-PCR)检测转染后细胞中miR-141表达水平,噻唑蓝(MTT)法检测SKOV3细胞增殖情况,流式细胞仪检测SKOV3细胞凋亡情况,Western blot检测NF-κB信号通路相关因子表达水平。添加NF-κB信号通路特异性抑制剂,观察SKOV3细胞对紫杉醇的敏感性变化。结果与Blank组和NC组比较,anti-miR-141组SKOV3细胞中miR-141的表达水平显著降低,差异有统计学意义(P<0.05),细胞增殖抑制率增加,细胞凋亡率升高,差异有统计学意义(P<0.05),NF-κB信号通路相关因子p-P65和p-IκBα的表达水平降低,差异有统计学意义(P<0.05)。NF-κB信号通路特异性抑制剂能够增强SKOV3细胞对紫杉醇的敏感性,差异有统计学意义(P<0.05)。结论转染miR-141抑制物后卵巢癌细胞SKOV3对紫杉醇敏感性增强,其作用机制可能与NF-κB信号通路的激活有关。Objective To investigate the effects of miR-141-mediated NF-κB signaling pathway on paclitaxel sensitivity in ovarian cancer-SKOV3 in vitro,and to explore its action mechanism.Methods The ovarian cancer SKOV3 cells that were transfected by the miR-141 inhibitor were enrolled as anti-miR-141 group,and the SKOV3 cells that were transfected by unrelated sequence were enrolled as NC group,moreover,the untransfected SKOV3 cells were enrolled as blank group.Real-time quantitative PCR(qRT-PCR)was used to detect the expression levels of miR-141 in transfected cells.The proliferation of SKOV3 cells was detected by MTT assay,and the apoptosis of SKOV3 cells was detected by flow cytometry.The expression levels of the factors related with NF-κB signal pathway were detected by Western Blot.And the specific inhibitors of NF-κB signaling pathway were added to observe the sensitivity of SKOV3 cells to paclitaxel.Results As compared with those in blank group and NC group,the expression levels of miR-141 were significantly decreased in anti-miR-141 group(P<0.05).The cell proliferation inhibition rate and apoptosis rate were significantly increased(P<0.05).Moreover the expression levels of NF-κB signaling pathway-related factors p-P65 and p-IκBαwere significantly decreased(P<0.05).Specific inhibitors of NF-κB signaling pathway could enhance the sensitivity of SKOV3 cells to paclitaxel(P<0.05).Conclusion The paclitaxel sensitivity in ovarian cancer-SKOV3 is increased after transfection of miR-141 inhibitor,and its action mechanism may be related to the activation of NF-κB signaling pathway.
