摘 要:目的:探究小檗碱对人卵巢癌上皮细胞HO-8910细胞株体外增殖的影响及机制。方法:CCK8法筛选小檗碱抑制HO-8910细胞株增殖的适宜浓度,细胞周期流式细胞法检测小檗碱对HO-8910细胞株G 1期和S期分布的影响,RT-PCR和Western blot法检测小檗碱对HO-8910细胞株增殖相关蛋白表达的影响。结果:50μmol/L小檗碱显著降低HO-8910细胞株活性,抑制HO-8910细胞株从G 1期进入S期,但对G 2期细胞分布无明显作用,小檗碱抑制HO-8910细胞株β-catenin和Cyclin D1 mRNA和蛋白表达,促进p21 mRNA和蛋白表达。结论:小檗碱通过抑制β-catenin和Cyclin D1,促进p21等增殖相关蛋白表达进而抑制HO-8910细胞株体外增殖。Objective:To explore the effect of berberine on the proliferation of human ovarian cancer cell line HO-8910 in vitro and its mechanism.Methods:CCK8 method was used to screen the suitable concentration of berberine to inhibit the proliferation of HO-8910 cell line.Cell cycle flow cytometry was used to detect the effect of berberine on the distribution of HO-8910 cell line in G 1 and S phases.RT-PCR and Western blot were used to detect the expression of proliferation related proteins of HO-8910 cell line treated by berberine.Results:50μmol/L berberine significantly reduced the activity of HO-8910 cell and inhibited HO-8910 from G 1 phase to S phase,but had no significant effect on the cell distribution of G 2 phase.Berberine inhibited the mRNA and protein expression ofβ-catenin and Cyclin D1 in HO-8910 cell line,and promoted p21 mRNA and protein expression.Conclusion:Berberine can inhibit the proliferation of HO-8910 cell in vitro by inhibiting the expression ofβ-catenin and Cyclin D1,promoting the expression of p21.
