摘 要:目的:通过大鼠正畸牙移动模型研究β-肾上腺素受体(β-adrenergic receptor,β-AR)阻滞剂对骨硬化蛋白(Sclerostin,SOST)和核因子κB受体活化因子配体(Receptor activator of nuclear factor kappa-B ligand,RANKL)的调控机制,以及对正畸牙移动的影响。方法:32只8周龄雄性Sprague-Dawley大鼠按照随机数字表法分为β_(1)-AR阻滞剂组(阿替洛尔)、β_2-AR阻滞剂组(布托沙明)、非选择性β-AR阻滞剂组(普萘洛尔)和对照组(生理盐水),每组8只。按体重每日灌胃给予相应药物或生理盐水,建立大鼠正畸牙移动模型并于4周后处死大鼠。通过游标卡尺测量牙移动距离,免疫组织化学染色观察张力侧和压力侧SOST和RANKL的表达情况并进行定量分析。结果:β-AR阻滞剂组大鼠的牙齿移动距离显著低于对照组,差异有统计学意义(P<0.05)。各组张力侧SOST和RANKL阳性细胞比例比较差异无统计学意义(P>0.05)。压力侧SOST和RANKL阳性细胞比例均为β-AR阻滞剂组低于对照组,差异有统计学意义(P<0.05)。结论:β-AR阻滞剂可减少正畸牙移动过程中压力侧SOST和RANKL的表达,并减小牙移动距离。Objective To explore the mechanism ofβ-adrenergic receptor blocker on sclerostin(SOST)and receptor activator of nuclear factor kappa-B ligand(RANKL),and its effect on orthodontic tooth movement by orthodontic tooth movement model in rats.Methods 328-week-old male Sprague-Dawley rats were randomly divided into theβ_(1)-AR blocker group(atenolol),theβ_(2)-AR blocker group(butoxamine),the non-selectiveβ-AR blocker group(propranolol)and the control group(physiological saline solution).Eight rats in each group were given blockers or physiological saline solution daily according to their body weight by intragastric administration.The model of orthodontic tooth movement was established and rats were killed 4 weeks later.The distance of tooth movement was measured by Vernier caliper,and the expression of SOST and RANKL on tension side and compression side were analysised by immunohistochemical staining.Results The distance of tooth movement in theβ-AR blocker groups was significantly lower than that in the control group,the differences were statistically significant(P<0.05).There was no significant difference in the proportion of SOST and RANKL positive cells on the tension side among the four groups(P>0.05).But theβ-AR blocker groups had significant lower proportion of SOST and RANKL positive cells on the compression side(P<0.05).Conclusionβ-AR blockers can reduce the expression of SOST and RANKL on the compression side and reduce the distance of tooth movement during orthodontic tooth movement.
