摘  要:为了探讨脂肪酸结合蛋白(adiposefatty acid binding protein,A-FABP)基因外显子1的多态性与北京油鸡、吉林黄鸡及其杂交F1代屠宰性能和肉品质的相关性,试验选择120~150日龄的吉林黄鸡自交亲本450只、北京油鸡自交亲本510只、北京油鸡♂×吉林黄鸡♀杂交F1代(正交)350只、吉林黄鸡♂×北京油鸡♀杂交F1代(反交)330只,均为雌雄各半,采用基因组PCR直接测序法筛选SNP位点,利用高分辨率熔解曲线技术(HRM)和竞争性等位基因特异性PCR法(KASP)进行多态性分析;同时测定各鸡群鸡只屠宰性能指标(活重、屠体率、半净膛率、全净膛率、胸肌率、腿肌率、腹脂率、翅膀重、消化道长度、肌胃重、脚重)和肉品质(胸肌和腿肌的pH_1值和pH_(24)值、滴水损失、失水率、熟肉率);最后将各基因型与屠宰性能和肉品质进行相关性分析。结果表明:在吉林黄鸡、北京油鸡及正反交F1代群体中的A-FABP基因外显子1中均存在C/T突变,且有CC、CT、TT 3个基因型;其中CC基因型在吉林黄鸡中为优势基因型,CT基因型在其他3个鸡群中为优势基因型,等位基因C在4个鸡群中均表现为优势等位基因;C/T突变在4个鸡群中均处于Hardy-Weinberg平衡状态,中度多态(0.25<PIC<0.5)且杂合度较低,有一定的遗传学意义。北京油鸡中CT基因型腿肌嫩度显著低于CC基因型(P<0.05),正交群体中CT基因型半净膛率与腿肌pH24值显著高于TT基因型(P<0.05),反交中CT基因型半净膛率显著高于CC基因型(P<0.05)。说明A-FABP基因外显子1的C/T突变可能是影响屠宰性状的潜在位点。In order to explore the correlation between the polymorphism of exon 1 of A-FABP gene and slaughter performance,meat quality of Beijing fatty chicken,Jilin yellow chicken and its hybrid F1 generation,450 self inbred parents of Jilin yellow chicken,510 self inbred parents of Beijing fatty chicken,330 Beijing fatty chicken♂×Jilin yellow chicken♀hybrid F1 generation(orthogonal),350 Jilin yellow chicken♂×Beijing fatty chicken♀hybrid F1 generation(backcross)were selected in the experiment,which all 120-150 days old and half male and half female.The SNP loci were screened by genomic PCR direct sequencing,and the polymorphisms were analyzed by high-resolution dissolution curve(HRM)and kompetitive allele specificity PCR(KASP).The slaughtering indexes(live weight,carcass rate,half only chamber rate,total only chamber rate,pectoral muscle ratio,leg muscle ratio,abdominal fat ratio,wing weight,digestive tract length,muscle stomach weight and foot weight)and meat quality(pH1 and pH24 values of chest and leg muscles,water loss,water loss rate and cooked meat rate)were measured;finally,the correlation between each genotype and slaughter performance,meat quality was analyzed.The results showed that there was a C/T mutation in exon 1 of A-FABP gene in Jilin yellow chicken,Beijing fatty chicken and reciprocal F1 population,and there were CC,CT and TT genotypes.Among them,CC was the dominant genotype in Jilin yellow chicken population,CT was the dominant genotype in the other three chicken populations,and C was the dominant gene in all four populations.C/T mutation in four populations was in Hardy-Weinberg equilibrium,moderate polymorphism(0.25<PIC<0.5)and low heterozygosity,which had certain genetic significance.The leg muscle shear force of CT genotype was significantly lower than that of CC genotype(P<0.05).The half only chamber rate and leg muscle pH24 value of CT genotype in orthogonal test were significantly higher than those of TT genotype(P<0.05).The half only chamber rate of CT genotype was significantly higher
