摘　　要：目的探讨长链非编码RNA(LncRNA)分化拮抗非蛋白质编码RNA(DANCR)的表达与非小细胞肺癌(NSCLC)患者铂类药物化疗敏感性及预后的关系。方法选取126例晚期NSCLC患者,采用荧光实时定量PCR(qRT-PCR)法测定LncRNA DANCR在NSCLC组织及其癌旁正常组织的表达,并分析其与化疗敏感性和预后的关系。结果NSCLC组织、癌旁正常组织中LncRNA DANCR的相对表达量分别为3.65±1.41、1.52±0.67,二者比较,P<0.01。癌组织中LncRNA DANCR表达与组织分化程度和TNM分期有关(P均<0.05)。化疗敏感与不敏感者LncRNA DANCR相对表达量分别为2.93±1.01、4.12±1.67,二者比较,P<0.01。LncRNA DANCR高表达者、低表达者的3年总生存率分别为12.31%、29.50%,二者比较,P<0.05。Cox回归分析显示,TNM分期Ⅳ期、低分化、铂类药物耐药及LncRNA DANCR高表达是影响NSCLC患者预后的独立危险因素(P均<0.05)。结论NSCLC组织中LncRNA DANCR呈高表达,且与组织分化程度、铂类药物耐药性和TNM分期有关,对评估预后有一定帮助,对化疗的敏感性有一定影响。Objective To investigate the relationships between the expression of long non-coding RNA(LncRNA)differentiation antagonizing non-protein coding RNA(DANCR)and the platinum drugs chemosensitivity and prognosis of patients with non-small-cell lung cancer(NSCLC).Methods Totally 126 patients with advanced NSCLC were select‐ed.Real-time quantitative PCR(qRT-PCR)was used to detect the expression of LncRNA DANCR in NSCLC tissues and their adjacent normal tissues,and their relationships with chemosensitivity and prognosis were analyzed.Results The relative expression of LncRNA DANCR in NSCLC tissues was 3.65±1.41,which was higher than that(1.52±0.67)in the adjacent normal tissues(P<0.01).The expression of LncRNA DANCR was related to the degree of differentiation and TNM stage(both P<0.05).The relative expression of LncRNA DANCR in chemotherapy sensitive patients was 2.93±1.01,which was lower than that(4.12±1.67)in chemotherapy insensitive patients(P<0.01).The 3-year overall sur‐vival rate in the high LncRNA DANCR expression group was lower than that in the low expression group(P<0.05).Cox regression analysis showed that TNM Ⅳ period,low differentiation,platinum drugs resistant and high expression of Ln‐cRNA DANCR were independent risk factors for the prognosis of NSCLC patients(all P<0.05).Conclusion LncRNA DANCR is highly expressed in NSCLC,and is related to the degree of differentiation and TNM stage and platinum drug re‐sistance,which might be helpful in evaluating the prognostic cycle and has a certain influence on the sensitivity of chemo‐therapy.
