摘  要:目的:探究肺力咳合剂调控核转录因子-κB(NF-κB)/丝裂原活化蛋白激酶(MAPK)信号通路对支气管哮喘大鼠气道炎症反应的影响。方法:取50只Wistar大鼠,其中40只腹腔注射卵清蛋白致敏和雾化吸入法建立哮喘大鼠模型,将其随机分为哮喘组、实验1组、实验2组、实验3组。剩余10只大鼠用生理盐水代替卵清蛋白干预,设为对照组。末次雾化后4 h,实验1组、实验2组、实验3组大鼠分别给予1 mL/kg、2 mL/kg、4 mL/kg肺力咳合剂灌胃,哮喘组和对照组均给予等量生理盐水灌胃,连续7 d。检测各组肺泡灌洗液(BALF)中炎症因子白介素(IL)-6、IL-1β、肿瘤坏死因子-α(TNF-α)水平,苏木精-伊红(HE)染色法观察各组肺组织病理学变化,RT-qPCR法检测肺组织NF-κB p65和p38 MAPK mRNA相对表达量,Western blotting法检测NF-κB磷酸化(p-)p65和p-p38 MAPK蛋白表达。结果:5组BALF中IL-6、IL-1β、TNF-α水平从低至高排列如下:对照组、实验2组、实验3组、实验1组、哮喘组,组间比较,差异均有统计学意义(均P<0.05);苏木精-伊红(HE)染色显示:哮喘组肺泡轮廓不清晰、肺泡壁水肿、增厚明显,多量炎性细胞浸润;3个实验组上述变化均较哮喘组减轻,其中实验2组减轻最为明显;各组大鼠肺组织中NF-κB p65、p38 MAPK mRNA相对表达量比较,差异无统计学意义(P>0.05);5组肺组织NF-κB p-p65/NF-κB p65、p-p38 MAPK/p38 MAPK蛋白比值从低至高排列如下:对照组、实验2组、实验3组、实验1组、哮喘组,组间比较,差异均有统计学意义(均P<0.05)。结论:肺力咳合剂可减轻哮喘大鼠气道炎症反应,其机制可能与抑制NF-κB、MAPK磷酸化激活有关。Objective:To investigatethe effect of Feilike mixture on airway inflammation in rats with bronchial asthma by regulating nuclear factor-κB(NF-κB)/mitogen-activated protein kinase(MAPK)signaling pathway.Methods:Forty Wistar rats were intraperitoneally injected with ovalbumin sensitization and atomized inhalation to establish the asthma model,and randomly divided into asthmatic group,experimental group 1,experimental group 2 and experimental group 3.Ten rats were treated with normal saline instead of ovalbumin and set as control group.Four hours after the last nebulization,the rats in experimental groups were given different doses of Feilike mixture,and the asthma group and the control group were given normal saline for 7 days.The levels of interleukin(IL)-6,IL-1β,tumor necrosis factor-α(TNF-α)in bronchial alveolar lavage fluid(BALF)were detected.The pathological changes of lung tissues were observed.The expressions of NF-κB p65 and p38 MAPK mRNA were determined by RT-qPCR.The NF-κB phosphorylated(p-)p65 and p-p38 MAPK protein expressions were measured by western blotting in lung tissues.Results:The levels of IL-6,IL-1β,TNF-αin BALF of the 5 groups were arranged from low to high as follows:control group,experimental group 2,experimental group 3,experimental group 1,asthma group.The differences among the 5 groups were statistically significant(P<0.05).Hematoxylin-eosin(HE)staining showed that the alveolar contour was not clear in the asthma group,the alveolar wall edema and thickening were obvious,and a large amount of inflammatory cell infiltration was observed.All the above changes in the three experimental groups were alleviated,and the improvement was most significant in experimental group 2.The expressions of NF-κB p65 and p38 MAPK mRNA were similar among groups(P>0.05).The ratio of NF-κB p-p65/NF-κB p65 and p-p38 MAPK/p38 MAPK was arranged from low to high as follows:control group,experimental group 2,experimental group 3,experimental group 1,asthma group,and the differences among groups were stati
