摘 要:珊瑚菜(Glehnia littoralis)又名北沙参,是一种常用药食同源植物。现珊瑚菜野生种群十分稀缺,是国家二级保护植物之一。本研究通过MISA软件对珊瑚菜转录组78828条Unigenes中简单重复序列(simple sequence repeats,SSR)位点信息进行挖掘与分析,共鉴定出14421个SSR位点,SSR发生频率为15.43%。珊瑚菜转录组SSR位点的种类从单碱基到六碱基重复均有分布,主要重复类型是单核苷酸和双核苷酸,分别占SSR总数的39.98%和40.98%。SSR位点的最小重复次数分布为5次,最大重复次数分布为24次,6和10次是SSR位点数量最多的重复次数,分别有3102和2672个,分别占总SSR数目的21.51%和18.53%。利用引物设计软件Primer 6.0软件,共设计出2894对引物。随机筛选出20对引物进行扩增验证,有12对引物扩增出片段。本研究结果表明珊瑚菜转录组SSR位点出现频率与密度较高且重复类型丰富,可为珊瑚菜遗传图谱的建立、遗传多样性的分析、物种的鉴定和育种等研究提供技术依据。Glehnia littoralis,also known as Beishashen,is a common medicine and food homologous plant.The existing wild population of G.littoralis is very scarce,which is one of the national second-class protected plants.In this study,MISA(MicroSatellite)was used to search and analysis of SSR from 78828 Unigenes of transcriptome data of G.littoralis.It was found that 14421 SSRs were identified in 78828 Unigenes,accounting for 15.43%of all the Unigenes.The species of SSR sites in the transcriptome of G.littoralis are distributed from single to six nucleotide.The mononucleotide and dinucleotide are the major repetitive types,accounting for 39.98%and 40.98%of all the SSRs,respectively.The distribution of minimum and maximum repeats of SSR loci was 5 and 24,of which 6 and 10 were the most,with 3102 and 2672 respectively,accounting for 21.51%and 18.53%of the total SSR.2894 pairs of primers were designed by Primer 6.0 software.20 pairs of primers were selected randomly for amplification and validation,and 12 pairs of primers amplified fragments.The results showed that the SSR loci in the transcriptome of G.littoralis had high frequency and density and rich repetition types,which provided technical basis for the construction of genetic map,analysis of genetic diversity,species identification and breeding of G.littoralis.
