摘 要:目的探讨非小细胞肺癌(NSCLC)患者外周血中微小RNA(miR)-25表达水平与病灶内癌基因表达变化的相关性。方法选择海南省人民医院2018年3月至2020年12月期间NSCLC患者为NSCLC组,以同期健康志愿者作为对照组,测定外周血中miR-25的表达水平以及肺癌组织、癌旁组织中miR-25、抑癌基因、增殖侵袭基因的表达水平,应用实时荧光定量聚合酶链反应(RT-qPCR)检测肺癌细胞株和正常肺上皮细胞BEAS-2B中的miR-25表达水平。不同组间计量资料的分析采用独立样本t检验。相关分析采用Pearson检验。结果NSCLC组患者外周血中miR-25的表达水平高于对照组(1.88±0.24比1.03±0.14,t=22.432,P<0.05),肺癌病灶内miR-25的表达水平高于癌旁病灶(2.19±0.32比1.05±0.17,t=21.797,P<0.05)且NSCLC组患者外周血中miR-25的表达水平与肺癌病灶内miR-25的表达水平呈正相关(r=0.641、P<0.01)。miR-25在NSCLC细胞株中的表达量高于正常人肺上皮细胞(2.05±0.35比1.01±0.15,t=15.237,P<0.05);肺癌病灶内Krüppel样因子4(KLF4)、B细胞易位基因2(BTG2)、含F-框及WD重复域蛋白7(FBXW7)、Kazal基序逆向诱导半胱氨酸丰富蛋白(RECK)的mRNA表达量水平均低于癌旁病灶(分别0.44±0.08比1.04±0.17、0.64±0.07比1.01±0.15、0.59±0.08比0.98±0.13、0.48±0.06比1.02±0.14,t=22.407、15.681、18.053、24.827,P<0.05)且与外周血中miR-25的表达水平呈负相关(r=-0.574、-0.625、-0.512、-0.663,P<0.05)。肺癌病灶内细胞周期蛋白(Cyclin)D1、c-Myc、N-钙黏蛋白(N-cadherin)、基质金属蛋白酶(MMP)-2、MMP-9的mRNA表达量水平均高于癌旁病灶(分别2.33±0.32比0.97±0.14、1.93±0.24比1.04±0.18、2.08±0.28比0.99±0.11、2.27±0.31比1.02±0.15、2.55±0.38比1.03±0.17,t=28.771、21.567、26.832、26.756、26.966,P<0.05)且与外周血中miR-25的表达水平呈正相关(r=0.564、0.692、0.503、0.672、0.485,P<0.05)。结论NSCLC患者外周血中miR-25的高表达与抑癌基因表达下调、增殖侵Objective To investigate the correlation between the expression of microRNA(miR)-25 in peripheral blood and the expression of oncogene in lesions of non-small cell lung cancer(NSCLC)patients.Methods The patients who underwent surgical treatment in Hainan Provincial People′s Hospital from March 2018 to December 2020 and were diagnosed with NSCLC were collected as the NSCLC group,and healthy volunteers who received physical examination in Hainan Provincial People′s Hospital during the same period served as the control group.The expression level of miR-25 in peripheral blood was determined,and the expression levels of miR-25,tumor suppressor genes,and proliferation and invasion genes in lung cancer lesions and adjacent lesions were examined.At the same time,RT-qPCR was used to detect the expression level of miR-25 in NSCLC cell lines and normal lung epithelial cells BEAS-2B.Measurement data between different groups were analyzed with independent sample t test.The Pearson test was used for the relevant analysis.Results The expression level of miR-25 in peripheral blood in NSCLC group was significantly higher than that in the control group(1.88±0.24 vs.1.03±0.14,t=22.432,P<0.05),the expression level of miR-25 in the lung cancer was significantly higher than that in the paracancerous lesions(2.19±0.32 vs.1.05±0.17,t=21.797,P<0.05),and the expression level of miR-25 in the peripheral blood in the NSCLC group was positively correlated with the expression level of miR-25 in the lung cancer lesions(r=0.641,P<0.01).The expression of miR-25 in NSCLC cell lines was significantly higher than that in normal human lung epithelial cells(2.05±0.35 vs.1.01±0.15,t=15.237,P<0.05).The mRNA expression levels of Kruppel-like factor 4(KLF4),B cell translocation gene 2(BTG2),F-box and WD repeat domain-containing 7(FBXW7),RECK in the lung cancer were significantly lower than those in the paracancerous lesions(0.44±0.08 vs.1.04±0.17,0.64±0.07 vs.1.01±0.15,0.59±0.08 vs.0.98±0.13,0.48±0.06 vs.1.02±0.14;t=22.407,15.681,18.053
