摘  要:目的探讨肌醇单磷酸酶域蛋白1(Inositol Monophosphatase Domain-Containing Protein 1,IMPAD1)基因高表达在食管癌中的功能和机制。方法对数生长期的食管癌细胞株Kyse70随机分为三组:空白组、IMPAD 1组与NC组。IMPAD1组在转染中加入pcDNA3.0-IMPAD1载体+lipofectamine 2000,NC组加入pcDNA3.0-NC载体+lipofectamine 2000,空白组加入lipofectamine 2000与等体积的生理盐水。CCK8法检测细胞增殖,伤口愈合实验检测细胞迁移,Transwell小室检测细胞侵袭,Western Blot检测蛋白表达。结果转染后24 h与48 h,IMPAD 1组的IMPAD1 mRNA表达水平高于空白组与NC组(P<0.05);细胞增殖指数高于空白组与NC组(P<0.05),细胞迁移与侵袭指数高于空白组与NC组(P<0.05),IMPAD1、磷脂酰肌醇3激酶(phosphatidylinositol 3 kinase,PI3K)蛋白相对表达水平高于空白组与NC组(P<0.05),空白组与NC组对比差异无统计学意义(P>0.05)。结论IMPAD1基因高表达能激活食管癌细胞的PI3K通路,从而促进食管癌细胞增殖、迁移与侵袭。Objective To investigate the function and mechanism of Inositol Monophosphatase Domain-Containing Protein 1(IMPAD1)gene overexpression in esophageal cancer.Methods The logarithmic growth phase of the esophageal cancer cell line Kyse70 were randomly divided into 3 groups:blank group,IMPAD1 group and NC group.In the IMPAD1 group,pcDNA3.0-IMPAD1 vector+lipofectamine 2000 were added to the transfection,the NC group were added pcDNA3.0-NC vector+lipofectamine 2000,and the blank group were added lipofectamine 2000 and equal volume of saline.The CCK8 method detects cell proliferation,the wound healing test detects cell migration,the Transwell chamber detects cell invasion,and Western Blot detects protein expression.Results At 24 h and 48 h after transfection,the IMPAD1 mRNA expression level in IMPAD1 group were higher than that of blank group and NC group(P<0.05);cell proliferation index were higher than that of blank group and NC group(P<0.05),cell migration and invasion index were higher than the blank group and NC group(P<0.05),the relative expression levels of IMPAD1 and phosphatidylinositol 3 kinase(PI3K)protein were higher than the blank group and NC group(P<0.05),and the comparision of the indexes between the blank group and NC group were not statistically significant(P>0.05).Conclusion The high expression of IMPAD1 gene can activate the PI3K pathway of esophageal cancer cells,thereby promoting the proliferation,migration and invasion of esophageal cancer cells.
