摘 要:目的:探讨稳定过表达三叶因子1(TFF1)对人肝细胞肝癌(HCC)细胞系MHCC97H增殖、侵袭、转移及EMT发生的影响。方法:使用TCGA数据库和qRT-PCR分析TFF1在人肝癌组织与正常组织中的表达情况。慢病毒介导TFF1感染MHCC97H细胞,获得稳定过表达TFF1的细胞株。Transwell实验检测细胞迁移和侵袭情况;CCK-8、集落形成实验测定细胞增殖和集落形成的变化。Western blot测定上皮-间质转化(EMT)标记性蛋白的表达情况。结果:生物信息分析和qRT-PCR检测显示TFF1在肝癌中低表达。利用慢病毒获得稳定过表达TFF1的MHCC97H细胞,过表达TFF1能够抑制细胞增殖、侵袭和迁移。EMT标记蛋白E-Cadherin在TFF1过表达组增高,Vimentin和N-Cadherin则下降,EMT进程因TFF1过表达而被抑制。结论:过表达TFF1抑制MHCC97H细胞的增殖、侵袭、迁移和EMT进程,其可能是HCC发生、发展的一种抑制因子。Objective:To explore the effects of trefoil factor 1(TFF1)overexpression on proliferation,invasion,migration and EMT of MHCC97H human hepatocellular carcinoma cells.Methods:TCGA database and qRT-PCR were used to analyze the expression of TFF1 in human hepatoma and normal tissues.TFF1 mediated by lentivirus was transfected into human hepatocellular carcinoma(HCC)cell line MHCC97H to generate stable cell lines.The cell migration and invasion were detected by Transwell test.In addition,cell proliferation and colony formation were measured by CCK-8 and colony formation respectively.The protein expressions of epithelial-mesenchymal transition(EMT)-related molecules were detected by Western blot.Results:Bioinformatics analysis and qRT-PCR results showed that TFF1 was poorly expressed in HCC.Stable overexpression of TFF1 was obtained in HCC cell line MHCC97H.Overexpression of TFF1 inhibited cell proliferation,invasion and migration.EMT-related protein E-cadherin increased in the TFF1 overexpression group,while Vimentin and N-cadherin decreased.Conclusion:Stable overexpression of TFF1 can inhibit proliferation,invasion,migration and EMT of MHCC97H hepatic cancer cell,which may be an inhibitor of hepatocellular carcinoma.
