摘 要:文章以转基因大穗小麦和春麦761幼苗叶片为实验材料,采用化学发光生物素探针标记Southern杂交法检测转基因大穗小麦中的特异性外源DNA片段,为进一步研究分析转基因大穗小麦中该特异性外源DNA片段奠定基础。结果表明,大赖草生物素标记探针与转基因大穗小麦酶切总DNA杂交有一条特异性杂交条带,片段长度约为750 bp~1000 bp,与春麦761酶切总DNA杂交后为空白,推测特异性条带所显示的外源性DNA片段来自大赖草总DNA。This paper takes transgenic spike wheat germplasm and seedling leaves of spring wheat 761 as experimental materials,and uses southern hybridization method,biotin-labeled probes,to detect specific exterior DNA fragments in transgenic wheat,which lays the foundation on further research and analysis in DNA fragment.The results show that there is a specific hybridization band between the biotin-labeled probe of Leymus chinensis and the total DNA digested with transgenic wheat.The fragment length is about 750 bp~1000 bp.After hybridization with the total DNA of spring wheat 761,it tends to be blank.It is speculated that the exogenous DNA fragment displayed by the specific band is from the total DNA of Leymus chinensis.
