摘  要:通过采用不同透明剂(柠檬烯、Van-Clear、正丁醇、松节油和二甲苯)制备石蜡切片,观察香菇(Lentinula edodes)子实体组织结构,确定最适透明剂,并利用优化石蜡切片方法对香菇子实体发育初期组织结构进行观察。结果表明,Van-Clear为最适透明剂,以其制备的石蜡切片中香菇子实体各部分组织结构清晰、完整、无皱缩变形。香菇菌盖皮层中的菌丝横向排列,菌丝间有大小不一的空隙,最外层菌丝排列紧密;菌肉中菌丝呈无规则致密扭结状态;菌褶中菌丝纵向规律排列,菌髓为同型结构;菌柄组织横切面呈无规则束状,存在不规律的小空隙结构。香菇原基内部菌丝呈致密无序交织状态;随着其不断膨大,下部左右两端分化出菌盖,并在菌盖下部出现栅栏状细胞整齐排列的子实层;随着菌盖的发育,菌褶结构逐渐形成。Different clearing agents were compared to improve the traditional paraffin section method and then the improved method was used to observe the tissue structure of Lentinula edodes during early fruiting body development.The results showed that the best clearing agent was Van-Clear,using which the tissue structure of each part of a fruiting body was clear,complete,and had no shrinkage or deformation.Pileus peels showed that hyphae in pileus cuticle were horizontally arranged with gaps of different sizes in between,and the outermost hyphae were tightly arranged.Context hyphae were irregularly densely kinked.Gill lamella hyphae were regularly arranged in a longitudinal manner and the hymenophoral trama was homoiomerous.The transverse section of stipe tissue was irregularly bundled with irregular small gaps in between.Primordial hyphae were dense and disorderly intertwined;and as they continuously expanded,t he the left and right ends of the lower part of the primordial hyphae differentiated into pileus with palisade cells neatly arranged at the bottom.As the development of the pileus structure continued,gill tissues gradually formed.
