摘 要:【目的】对比分析木薯花芽分化株和花芽未分化株的内源激素和氨基酸含量,为木薯成花机理研究及花期调控提供理论依据。【方法】采用液相色谱—质谱联用(LC-MS)技术测定木薯品种新选048花芽分化株和花芽未分化株地上部茎、叶和芽的15种内源激素含量和23种氨基酸含量,分析其差异性和相关性。【结果】与新选048木薯花芽未分化株相比,花芽分化株茎的赤霉素4(GA_(4))、赤霉素7(GA_(7))、玉米素核苷(ZR)、反玉米素核苷(TZR)、水杨酸(SA)、腺嘌呤(Adenine)和腺苷(Adenosine)含量及D-α-氨基正丁酸(RABA)、甘氨酸(Gly)、谷氨酰胺(Gln)、甲硫氨酸(Met)、酪氨酸(Tyr)、丝氨酸(Ser)、酪氨酸(Thr)、天冬酰胺(Asn)、缬氨酸(Val)、异亮氨酸(ILE)含量极显著升高(P<0.01,下同),生长素(IAA)、油菜素内酯(BR)和赤霉素1(GA_1)含量显著(P<0.05,下同)或极显著降低,其中,GA_(4)、ZR、SA、Adenosine和Adenine含量差异在3倍以上,Gln、Met、Thr、Tyr、Val和ILE含量差异在6倍以上;各内源激素含量均与部分氨基酸(3种或3种以上)含量呈显著或极显著相关,除ZR含量与Met含量外,赤霉素3(GA_(3))、GA_(4)、GA_(7)、ZR、TZR、Adenine和Adenosine含量与Gln、Met、Tyr、Thr、Val、ILE和组氨酸(His)含量均呈显著或极显著正相关,IAA和BR含量与Gln、Met、Tyr、Thr、Val、ILE和His含量呈显著或极显著负相关。【结论】木薯从营养生长过渡到花芽分化的生殖生长,其茎部的内源激素和氨基酸反应活跃,其中,GA_(4)、GA_(7)、ZR、TZR、SA、Adenine、Adenosine、RABA、Gly、Gln、Met、Tyr、Ser、Thr、Asn、Val和ILE等的累积,以及IAA、BR和GA_1等的降解对其花芽分化具有促进作用;GA_(4)、Adenosine、Adenine、Gln、Met、Thr、Tyr、Val和ILE含量大幅度升高是指示木薯花芽分化起始的最佳生理指标。【Objective】The contents of endogenous hormones and amino acids in cassava flower bud differentiated and flower bud undifferentiated plants were compared to study the flowering mechanism and further provide the regulation of cassava flowering period.【Method】Contents of 15 endogenous hormones and 23 amino acids in stalk,leaf and bud of flower bud differentiated plant and flower bud undifferentiated plant of cassava variety Xinxuan 048 were tested by LC-MS and analyzed.【Result】Compared with flower bud undifferentiated plant of cassava variety Xinxuan 048,gibberellin 4(GA_(4)),gibberellin 7(GA_(7)),ribosylzeatin(ZR),trans-zeatin riboside(TZR),salicylic acid(SA),adenine,adenosine,D-α-aminobutyric acid(RABA),glycocoll(Gly),glutamine(Gln),methionine(Met),tyrosine(Tyr),serine(Ser),threonine(Thr),asparagine(Asn),valine(Val)and isoleucine(ILE)in stalk of flower bud differentiated plant increased extremely significantly(P<0.01,the same as below),while auxin(IAA),brassinolide(BR)and gibberellin 1(GA1)decreased significantly(P<0.05,the same as below)or extremely.Among then,the content difference of GA_(4),ZR,SA,adenosine and adenine were more than 3 times,while Gln,Met,Thr,Tyr,Val and Ile were more than 6 times.Each endogenous hormone was significantly or extremely significantly correlated with at least 3 amino acids.Beside ZR and Met,gibberellin 3(GA_(3)),GA_(4),GA_(7),ZR,TZR,adenine,adenosine were significantly or extremely significantly positively correlated with Gln,Met,Tyr,Thr,Val,ILE,histidine(His).While IAA and BR were significantly or very significantly negatively correlated with those amino acids.【Conclusion】In the transition from vegetative growth to reproductive growth,represented by the growth of flower bud differentiation,endogenous hormones and amino acids in cassava stalk were mostly active.The accumulation of GA_(4),GA_(7),ZR,TZR,SA,adenine,adenosine,RABA,Gly,Gln,Met,Tyr,Ser,Thr,Asn,Val,ILE,and the degradation of IAA,BR,GA1 are stimulative for the flower bud differentiation.The content of GA_(
