食管癌组织lncRNA PCAT-1、ANXA10表达变化及其临床意义

摘　　要：目的观察食管癌组织长链非编码RNA(lncRNA)前列腺癌相关转录因子1(PCAT-1)、膜联蛋白A10(ANXA10)表达变化,并探讨其临床意义。方法选择食管癌患者87例,取手术切除的食管癌组织及其配对的癌旁正常组织,采用实时荧光定量PCR技术检测lncRNA PCAT-1、ANXA10表达,比较食管癌组织与癌旁正常组织lncRNA PCAT-1、ANXA10表达差异。收集食管癌患者临床病理资料,分析lncRNA PCAT-1、ANXA10表达与患者临床病理特征的关系。以食管癌组织lncRNA PCAT-1、ANXA10相对表达量的中位数为临界值,将患者分为lncRNA PCAT-1高表达者与lncRNA PCAT-1低表达者、ANXA10高表达者与ANXA10低表达者。术后随访3年,比较不同lncRNA PCAT-1、ANXA10表达者3年总体生存率差异。结果食管癌组织lncRNA PCAT-1相对表达量高于癌旁正常组织,ANXA10相对表达量低于癌旁正常组织(P均<0.05)。食管癌组织lncRNA PCAT-1、ANXA10相对表达量均与肿瘤TNM分期、淋巴结转移有关(P均<0.05),而与性别、年龄、酗酒、肿瘤直径、组织分化程度无关(P均>0.05)。食管癌组织lncRNA PCAT-1表达与ANXA10表达呈负相关关系(r=-0.757,P<0.05)。lncRNA PCAT-1高表达者43例、lncRNA PCAT-1低表达者44例,lncRNA PCAT-1高表达者3年总体生存率低于lncRNA PCAT-1低表达者(χ^(2)=4.167,P<0.05);ANXA10高表达者41例、ANXA10低表达者46例,ANXA10高表达者3年总体生存率高于ANXA10低表达者(χ^(2)=5.696,P<0.05)。结论食管癌组织lncRNA PCAT-1高表达、ANXA10低表达,二者表达与肿瘤TNM分期、淋巴结转移和患者预后有关。lncRNA PCAT-1、ANXA10有可能成为食管癌早期诊断、靶向治疗和预后评估的生物标志物。Objective To observe the expression changes of long non-coding RNA(lncRNA)prostate cancer-associ⁃ated transcription factor-1(PCAT-1)and annexin A10(ANXA10)in the esophageal cancer tissues,and to explore their clinical significance.Methods Eighty-seven patients with esophageal cancer were selected,and the resected esopha⁃geal cancer tissues and the matched adjacent normal tissues were obtained.The expression levels of lncRNA PCAT-1 and ANXA10 were detected by real-time quantitative PCR,and the differences in the expression levels of lncRNA PCAT-1 and ANXA10 were compared between the esophageal cancer tissues and adjacent normal tissues.The clinicopathological data of patients with esophageal cancer were collected,and the relationship between the expression of lncRNA PCAT-1 and ANXA10 and the clinicopathological characteristics of patients with esophageal cancer was analyzed.The median rel⁃ative expression of lncRNA PCAT-1 and ANXA10 in the esophageal cancer was used as the critical value,and then the patients were divided into the high lncRNA PCAT-1 expression group and low lncRNA PCAT-1 expression group,and the high ANXA10 expression group and low ANXA10 expression group.The patients were followed up for 3 years,and the 3-year overall survival rates of patients with different lncRNA PCAT-1 and ANXA10 expression levels were com⁃pared.Results The relative expression of lncRNA PCAT-1 in the esophageal cancer tissues was higher than that in the adjacent normal tissues,and the relative expression of ANXA10 was lower than that in the adjacent normal tissues(both P<0.05).The relative expression levels of lncRNA PCAT-1 and ANXA10 in the esophageal cancer were related to TNM stage and lymph node metastasis(all P<0.05),but not related to gender,age,alcoholism,tumor diameter,or degree of tissue differentiation(all P>0.05).The expression of lncRNA PCAT-1 was negatively correlated with the expression of ANXA10 in esophageal cancer(r=-0.757,P<0.05).There were 43 cases of high expression of lncRNA PCAT-1 and 44 cases of
