摘 要:水体氨氮(NH_(4)^(+)-N)和硝氮(NO_(3)^(–)-N)的测定是浙江大学生态学本科实验教学中的重要内容之一。该实验利用紫外分光光度计与CleverChem Anna全自动间断化学分析仪对水体中的NH_(4)^(+)-N和NO_(3)^(–)-N进行测定。为了对比分析2种仪器在样品测定中的准确度、手动稀释和仪器自动稀释造成的差异以及由不同稀释倍数造成的数据误差等问题,该文利用2种仪器对NH_(4)^(+)-N和NO_(3)^(–)-N标准使用液(1、10、100 mg/L)的测定结果进行分析。结果表明:2种仪器的测定结果虽有一定的差异但具有较高的相关性(NO_(3)^(–)-N,R^(2)=0.997;NH_(4)^(+)-N,R^(2)=0.996),可通过校正曲线进行校正。数据存在差异的原因可能是仪器自动稀释和样品中干扰因素的影响。通过对2种教学仪器在水体NH_(4)^(+)-N和NO_(3)^(–)-N的测定结果分析,加深了学生对测定原理的理解,提升学生实验过程中的思考能力,以期为相关的实验教学提供参考。The measurement of ammonia nitrogen and nitrate nitrogen in water is one of the important contents in the undergraduate experiment teaching of ecology in Zhejiang University.The ultraviolet spectrophotometer and automatic intermittent chemical analyzer are used to determine the NH_(4)^(+)-N and NO_(3)^(–)-N in water.The standard samples(1、10、100 mg/L)of NH_(4)^(+)-N and NO_(3)^(–)-N for analysis are adopted to compare the accuracy of the two instruments,the difference caused by manual dilution and automatic dilution of the instrument,and the data error caused by different dilution ratios and so on.The results show that although there are some differences between the results of the two instruments,there is a high correlation(NO_(3)^(–)-N,R^(2)=0.997;NH_(4)^(+)-N,R^(2)=0.996),which can be corrected through the correction curve.The difference of data may be caused by the influence of the automatic dilution of the instruments and the interference factors in the samples.Through the analysis of the measurement results of NH_(4)^(+)-N and NO_(3)^(–)-N in water by two kinds of teaching instruments,the students'understanding of the measurement principle is deepened,and their thinking ability in the process of experiment is improved,so as to provide reference for relevant experimental teaching.
