摘  要:目的:通过网络药理学的方法探究茶叶抗癌的作用机制,为揭示茶叶抗癌的科学内涵提供新思路。方法:通过中国知网进行文献研究,收集茶叶中主要的化学成分;利用化源网检索进一步确定各个化学成分的化学结构式,并使用ChemBioDraw软件手动画取;将取得的化学结构式导入SwissTargetPrediction数据库预测成分的作用靶点;利用Genecards和Drugbank两个数据库获取癌症相关靶点;通过String数据库构建PPI网络,并根据Degree值的大小筛选核心靶点;采用Metascape数据库对核心靶点进行GO及KEGG富集分析。结果:通过文献研究最终纳入茶叶中13个化学成分,共预测出165个作用靶点;纳入癌症相关靶点3350个;通过PPI确认IL6、VEGFA、TNF等40个核心靶点;GO生物功能富集分析发现,积极调控细胞迁移、调节平滑肌细胞增殖、对氧气水平的反应等生物过程;膜筏、细胞质核周区域、转录因子复合体等细胞组成;细胞因子受体结合、内肽酶活性、蛋白磷酸酶结合等分子作用;KEGG信号通路结果显示,主要涉及多种癌症的相关通路、PI3K-Akt信号通路和NF-κB信号通路等。结论:茶叶中儿茶素及其酯类衍生物可能是起抗癌作用的主要成分,多成分多靶点的综合调控是茶叶发挥药效物质基础的主要优势。To explore the anti-cancer mechanism of tea by means of network pharmacology,and provide new ideas for revealing the scientific connotation of tea anti-cancer.Methods:Document research through China HowNet to collect the main chemical components in tea;use the chemical source network to search and further determine the chemical structure formulas of each chemical component,and use ChemBioDraw software to draw it manually;import the obtained chemical structure into the SwissTargetPrediction database to predict the target of the components;use two databases of Genecards and Drugbank to obtain cancer-related targets;construct a PPI network through the String database,and screen core targets according to the Degree value;use Metascape database to perform GO and KEGG enrichment analysis on core targets.Results:Through literature research,13 chemical components in tea were finally included,and a total of 165 targets were predicted;3,350 cancer-related targets were included;40 core targets including IL6,VEGFA,and TNF were confirmed by PPI;GO biological function enrichment analysis found that it actively regulated cell migration,smooth muscle cell proliferation,response to oxygen levels and other biological processes;membrane rafts,cytoplasmic perinuclear regions,transcription factor complexes and other cellular components;cytokine receptor binding,endopeptidase activity,protein phosphatase binding and other molecular effects;The results of KEGG signaling pathway showed that it mainly involved various cancer-related pathways,PI3K-Akt signaling pathway,NF-κB signaling pathway and so on.Conclusion:The catechins and their ester derivatives in tea may be the main components with anti-cancer effects,and the comprehensive regulation of multiple components and multiple targets is the main advantage of the material basis of tea's efficacy.
