摘 要:目的观察吗啡暴露对猫行为敏化及其中枢神经系统神经元的影响,以为临床上更好地治疗吗啡依赖性提供行为学及病理学依据。方法将12只健康家猫随机分成对照组3只和实验组9只。实验组猫于每日8点、12点、16点皮下注射盐酸吗啡,药量逐日递增,连续5 d;对照组猫在同样的时间和部位注射等量的生理盐水。停药3 d后,于第9天评定2组猫的行为敏化情况(扭体、湿狗样抖、站立、跳跃、清理皮毛、齿颤、上睑下垂次数);3周后在电镜下观察2组猫各脑区皮质神经元的超微结构。结果实验组猫发生扭体、湿狗样抖、站立、跳跃、清理皮毛、齿颤、上睑下垂的次数均明显多于对照组(P均<0.05),存在行为敏化现象。电镜下观察,实验组猫皮质神经元均发生不同程度的改变,包括凋亡、变性、坏死,细胞器减少、线粒体肿胀、染色质边集、核固缩甚至坏死。结论吗啡可导致多种脑组织病理损害,吗啡成瘾猫的行为敏化与脑神经元凋亡有密切关系。Objective It is to observe the effect of morphine exposure on cat behavioral sensitization and its central nervous system neurons,so as to provide behavioral and pathological evidences for better treatment of morphine dependence in clinical practice.Methods Twelve healthy domestic cats were randomly divided into experiment group(n=9)and control group(n=3).The cats in the experimental group were injected subcutaneously with morphine hydrochloride at 8 o’clock,12 o’clock,and 16 o’clock every day,and the dose was increased daily,treated for 5 days;the cats in the control group were injected with the same amount of normal saline at the same time and position.In 3 days after stopping the drug,the behavioral sensitization(writhing,wet dog shaking,standing,jumping,cleaning the fur,tooth tremor,and ptosis)was assessed on the 9th day;3 weeks later,the ultrastructure of cortical neurons in each brain area of the cats of the two groups were observed under the electron microscope.Results The cats in the experimental group with behavioral sensitization had more times of writhing,wet dog-like shaking,standing,jumping,cleaning fur,tooth shaking,and ptosis than ones in the control group(all P<0.05).The electron microscope showed that the cortical neurons of the rats in the experimental group had different degrees of changes,including apoptosis,degeneration,necrosis,reduction of organelles,mitochondrial swelling,chromatin margination,nuclear pyknosis and even necrosis.Conclusion Morphine can cause a variety of brain tissue pathological damages.The behavioral sensitization of morphine-addicted cats is closely related to brain neuron apoptosis.
