摘  要:旨在探究猪源沙门氏菌生物被膜形成能力(成膜能力)对其耐药性和毒力的影响。通过96孔微量板法、K-B纸片法、微量稀释法和寇氏改良法,测定来源于腹泻仔猪的69株沙门氏菌的成膜能力、耐药表型、最小抑菌浓度(MIC)和对小鼠的半数致死量(LD_(50))。结果显示,69株沙门氏菌成膜阳性率为92.8%(64/69),以弱阳性(1+)为主(84.4%,54/64)。成膜菌对四环素(TET)和强力霉素(DOX)的耐药率最高,分别为76.6%(49/64)和75.0%(48/64);多重耐药率为15.6%(10/64),共有9种多重耐药谱,耐药模式70.0%为四环素-强力霉素-阿莫西林-氨苄西林-甲氧苄啶-复方新诺明-氟苯尼考(TET-DOX-AMX-AMP-TMP-SXT-FFC);TET和DOX对成膜菌的MIC较其浮游菌分别提高2倍~16倍和2倍~8倍。强阳性(3+)成膜菌对小鼠的LD_(50)值最大,与弱阳性(1+)之间差异显著(P<0.05)。表明受试沙门氏菌普遍具有成膜能力,多重耐药范围广、耐药谱复杂,成膜菌对TET和DOX的耐药性与其成膜能力呈正相关,随着成膜能力增强,其LD_(50)值也随之增大。To investigate the effect of biofilm formation(film-forming ability)on drug resistance and virulence of porcine-derived Salmonella,the 96-well microplate method,the Kirby-Bauer disk diffusion method,the microdilution method and the improved Karber method were used to determine the biofilm forming ability in vitro,drug-resistant phenotype,minimum inhibitory concentrations(MICs)of biofilm bacteria and planktonic bacteria and median lethal dose(LD50)in mice on 69 strains of Salmonella from diarrhea piglets respectively.The results showed that the positive rate of biofilm formation was 92.8%(64/69)in 69 strains of Salmonella,which were mainly weak positive(84.4%,54/64).The resistance rates of TET and DOX were the highest for membrane-forming bacteria,which were 76.6%(49/64)and 75.0%(48/64),respectively.The multidrug resistance rate is 15.6%(10/64),showing a total of 9 multi-drug resistance spectra,with 70.0%of the drug resistance patterns being TET-DOX-AMX-AMP-TMP-SXT-FFC.The MICs of ciprofloxacin(CIP)and florfenicol(FFC)on biofilm bacteria was increased by 2-16 times and 2-8 times than that of the corresponding planktonic bacteria,respectively.The LD_(50) of strong positive biofilm bacteria(3+)was the highest,and the difference was significantly different from that of the weak positive biofilm bacteria(1+)(P<0.05).The tested Salmonella had near-universal biofilm forming ability,the multidrug resistance range is broad and the drug resistance spectrum is complex.The drug resistance of biofilm bacteria to ciprofloxacin(CIP)and florfenicol(FFC)was positively correlated with biofilm forming ability,and the LD50 increased with the enhancement of biofilm forming ability.
