摘 要:为了分析宠物猫细小病毒(FPV)流行株的VP2基因的遗传变异情况,通过病毒分离、形态学观察、PCR方法及间接免疫荧光试验(IFA)从疑似FPV感染的宠物猫粪便和血清等病料中分离得到1株FPV,命名为XNF-1株。采用人工感染试验检测其XNF-1株的致病性,采用PCR方法扩增XNF-1株VP2基因并测序,分析其遗传变异情况。结果显示,XNF-1株对猫具有致病性,XNF-1株与16株参考株VP2基因同源性在98.1%〜99.9%之间,与FPV-C株、FPV-D株、FPV-A株位于同一分支,其中与FPV-A株位于同一分支.遗传距离最近,与其他参考株遗传距离较远。研究结果为宠物猫细小病毒病防控及疫苗研究奠定了基础。In order to analyze the genetic variation of VP2 gene of pet cat parvovirus(FPV)epidemic strains,one strain of FPV,named XNF-1 strain,was isolated from feces and serum of suspected pet cat FPV by virus isolation,morphological observation,PCR and IFA.The results showed that XNF-1 strain had pathogenicity to cats.The VP2 gene homology between XNF-1 and 16 reference strains ranged from 98.1%to 99.9%.XNF-1 is located in the same branch with FPV-C,FPV-D and FPV-A strains.Among them,XNF-1 is located in the same branch with FPV-A strain and has the closest genetic distance with other reference strains.This study laid a foundation for the prevention and control of pet cat parvovirus disease and vaccine research.
