摘 要:目的探究解毒祛瘀滋阴方(Jiedu Quyu Ziyin Recipe,JQZR)对刀豆蛋白A(concanavalin A,Con A)诱导的自身免疫性肝炎(autoimmune hepatitis,AIH)小鼠肝损伤的保护作用.方法40只SPF级C57BL/6小鼠随机分为5组(n=8):对照组、模型组、模型+JQZR(5、10、20 g/kg)组.对照组常规饲养,模型组尾静脉注射15 mg/kg Con A,模型+JQZR各组分别给小鼠灌胃JQZR 3 d后尾静脉注射15 mg/kg Con A,各组小鼠均在注射Con A 12 h后处死,收集血清.全自动生化分析仪分析小鼠血清中肝功能指标和血脂代谢生化指标;HE染色观察小鼠肝组织损伤情况;TUNEL染色观察肝组织凋亡;ELISA检测外周血中炎性因子水平;WB检测肝组织中凋亡相关蛋白、iNOS和IL-10蛋白表达;免疫组化检测肝组织中NF-κB的含量.结果与对照组比较,模型组小鼠肝功能受损,血脂代谢异常;肝组织炎性细胞浸润,肝窦结构紊乱,细胞凋亡率增加(P<0.05),炎性因子水平升高(P<0.05),NF-κB表达上调(P<0.05).与模型组比较,JQZR10、20 g/kg明显明显改善肝功能和血脂代谢紊乱(P<0.05),并有效减轻肝组织损伤,减少细胞凋亡(P<0.05),减轻肝组织炎症并下调NF-κB的表达(P<0.05).结论JQZR通过抗炎和抗凋亡明显改善Con A诱导的小鼠急性肝损伤.Objective To investigate the effects of Jiedu Quyu Ziyin Recipe(JQZR)on liver injury in mice with autoimmune hepatitis(AIH)induced by concanavalin A(Con A).Methods Forty SPF C57BL/6 mice were randomly divided into 5 groups(n=8):control group,model group,3 model+JQZR groups(5,10,20 g/kg).The control group was fed routinely.The mice in the model group were injected with 15 mg/kg Con A via the tail vein,and those in the model+JQZR groups were intragastrically given JQZR for 3 days before injection of 15 mg/kg Con A through the tail vein.The mice were sacrificed 12 hours after injection with Con A and sera were collected.The automatic biochemical analyzer was used to analyze liver function and the levels of lipid metabolism indicators in the sera of mice.HE staining was performed to observe the liver tissue damage,TUNEL staining to observe the apoptosis of liver tissues,ELISA to detect the levels of inflammatory factors in peripheral blood,and Western blotting to detect the expression of apoptosis-related protein,iNOS and IL-10 in liver tissue.The expression of NF-kB in liver tissue was immunohisto-chemically detected.Results In the model group,the liver function was impaired and lipid metabolism was abnormal.The inflammatory cells infiltrated in liver tissue.The structure of hepatic sinuses was disorganized.The apoptosis rate was significantly increased(P<0.05),the level of inflammatory factors was significantly enhanced(P<0.05),and the expression of NF-kB was significantly up-regulated(P<0.05).JQZR(10 and 20 g/kg)could significantly improve liver function and dyslipidemia(P<0.05),effectively reduce liver tissue injury and apoptosis(P<0.05),alleviate liver tissue inflammation and down-regulate NF-κB expression(P<0.05).Conclusion JQZR can significantly improve acute liver injury induced by Con A by anti-inflammatory and anti-apoptotic effects.
