摘  要:目的探究亚硒酸钠对肺癌细胞生物学行为的影响以及如何通过NF-κB信号通路影响肺癌凋亡的具体机制。方法CCK-8细胞增殖实验、细胞划痕实验观察肺癌细胞的增殖和迁移的能力;流式细胞术、qPCR和Western blot实验检测不同浓度亚硒酸钠处理后的肺癌细胞的凋亡率、凋亡标志因子Bcl-2和Bax的mRNA和NF-κB信号通路相关蛋白(NF-κB、p-NF-κB、IkBα、p-IkBα)的变化水平;细胞免疫荧光实验观察p-NF-κB在细胞内的变化水平和分布情况。结果CCK-8细胞增殖实验和细胞划痕实验显示肺癌细胞增殖和迁移被亚硒酸钠显著抑制。流式结果显示,亚硒酸钠处理后细胞凋亡率明显增加。与对照组相比,亚硒酸钠组的p-NF-κB、p-IkBα、Bcl-2蛋白的表达水平和Bcl-2 mRNA表达水平均下调,而Bax蛋白和mRNA表达水平则与之相反。细胞免疫荧光实验结果显示亚硒酸钠可使NF-κB的磷酸化水平受到抑制。qPCR结果显示,亚硒酸钠组与BAY11-7082(NF-κB抑制剂)均可使Bcl-2 mRNA表达水平下调,Bax mRNA表达水平则上调。结论亚硒酸钠可能抑制肺癌细胞的增殖和迁移,同时通过抑制NF-κB信号通路诱导肺癌细胞的凋亡。Objective To investigate the effect of sodium selenite on the biological behavior of lung cancer cells and the specific mechanism of its effect on lung cancer apoptosis through the NF-κB signaling pathway.Methods CCK-8 cell proliferation assay and cell scratch assay were used to observe the proliferation and migration of lung cancer cells.Flow cytometry,qPCR and Western blot were used to detect the apoptosis of lung cancer cells and the expression of Bcl-2,Bax,NF-κB,p-NF-κB,IkBα,p-IkBαin lung cancer cells respectively.The distribution of p-NF-κB in lung cells was observed by immunofluorescence assay.Results The results of CCK-8 cell proliferation assay and cell scratch assay showed that lung cancer cell proliferation and migration were significantly inhibited by sodium selenite.The results of Flow cytometry showed that the apoptosis of lung cancer cells significantly increased after sodium selenium treatment.Compared with the control group,the expression levels of p-NF-κB,p-IkBα,Bcl-2 protein and Bcl-2 mRNA in the sodium selenite group were down-regulated,while the expression of Bax protein and mRNA were up-regulated.The results of immunofluorescence experiments showed that sodium selenite could inhibit the phosphorylation level of NF-κB.The qPCR results showed that sodium selenite and BAY11-7082(NF-κB inhibitor)could down-regulate the expression level of Bcl-2 mRNA,while up-regulate the expression of Bax mRNA.Conclusion Sodium selenite may inhibit proliferation and migration of lung cancer cells and induce apoptosis by inhibiting the NF-κB signaling pathway.
