摘 要:长链非编码RNA(lncRNA)参与肝脏的发育、代谢、纤维化、癌变,控制肝脏从发育到稳态再到病态的关键进程。肝星状细胞(HSC)是肝纤维化的中心细胞,而lncRNA能调控HSC的活化增殖、凋亡、自噬等,把控肝纤维化相关信号通路的启动。lncRNA不仅贯穿了HSC的活化、凋亡、自噬等环节,促使各环节之间相互影响,且还与微RNA(miRNA)以竞争性的关系构成了一个复杂的调控网络,影响着肝纤维化的进展。故lncRNA可作为治疗靶点应用于抗肝纤维化或以标志物的形式预测肝纤维化的进展。未来,深入研究lncRNA位点功能和lncRNA、miRNA、下游靶基因之间的调控机制,有望发现肝纤维化治疗的新靶点。Long non-coding RNA(lncRNA)is involved in the development,metabolism,fibrosis and carcinogenesis of liver,and controls the key process of liver from development to homeostasis,and then to pathology.Hepatic stellate cells(HSC)are the central cells of hepatic fibrosis,and lncRNA can regulate the activation,proliferation,apoptosis and autophagy of HSC,and control the activation of hepatic fibrosis related signaling pathways.LncRNA not only plays an important role in HSC activation,apoptosis and autophagy,but also forms a complex regulatory network with microRNA(miRNA)competitively,which affects the progress of hepatic fibrosis.Therefore,lncRNA can be used as a therapeutic target in the treatment of liver fibrosis or as a marker to predict the progress of hepatic fibrosis.In the future,further study on the function of lncRNA sites and the regulatory mechanism between lncRNA,miRNA and downstream target genes is expected to discover new targets for the treatment of hepatic fibrosis.
