肺腺癌中TNF-α、SOD-2与增殖的关系研究

摘　　要：目的分析TNF-α、SOD-2与增殖相关蛋白CyclinD1和Survivin在肺腺癌组织中的表达及各指标之间的关系,并探讨SOD-2与肺腺癌患者临床病理特征和生存预后的关系。方法收集河北医科大学第二医院病理科2009年1月至2014年12月诊断的68例肺腺癌标本,免疫组织化学方法检测TNF-α、SOD-2和增殖相关蛋白CyclinD1、Survivin的表达;统计上述各指标之间的相关性;回顾性研究SOD-2表达和患者临床病理特征、生存及预后的关系。结果肺腺癌组织中TNF-α、SOD-2、Survivin和CyclinD1均表达升高,且TNF-α和SOD-2,SOD-2和Survivin及CyclinD1的表达之间均存在正相关关系。SOD-2表达阳性肺腺癌患者更易发生淋巴结转移,且生存时间明显降低。结论在人肺腺癌标本中,炎症细胞因子TNF-α表达升高,其可能通过促进SOD-2表达介导肺腺癌细胞增殖。SOD-2高表达和肺腺癌患者的不良生存预后相关,可以作为预测预后的独立指标。Objective To investigate the expression of TNF-α,SOD-2,CyclinD1 and Survivin in lung adenocarcinoma and their correlation,and to explore the relationship between SOD-2 and pathological characteristics as well as survival prognosis of lung adenocarcinoma.Methods A total of 68 lung adenocarcinoma specimens were collected from department of pathology of our hospital from January 2009 to December 2014.The expression levels of TNF-α,SOD-2,CyclinD1 and Survivin were detected by immunohistochemistry.The correlation between the above indexes was analyzed.And the correlation between SOD-2 expression and clinicopathological features,survival and prognosis was retrospectively analyzed.Results The expression levels of TNF-α,SOD-2,survivin and CyclinD1 were increased in lung adenocarcinoma tissues,and there was a positive correlation between the expression levels of TNF-αand SOD-2,between SOD-2 and Survivin as well as CyclinD1.Moreover the patients with high SOD-2 expression were more likely to have lymph node metastasis,and the survival time was significantly lower than that of patients with low SOD-2 expression.Conclusion The high expression of TNF-αin human lung adenocarcinoma speciments may mediate the proliferation of lung adenocarcinoma cells by promoting the expression of SOD-2.Moreover the high expression of SOD-2 is associated with poor survival and prognosis of patients with lung adenocarcinoma,which can be used as an independent prognostic indicator.
