唾液细胞外囊泡miRNA辅助口腔鳞癌诊断与术后评估

摘　　要：目的分析唾液源细胞外囊泡中miRNA在OSCC诊断及术后评估中的意义。方法分别比较17例OSCC患者与11例健康人群的唾液细胞外囊泡总miRNA的表达差异;6例OSCC患者术前血液与唾液细胞外囊泡中miR-21-5p的表达差异;7例OSCC患者术前与术后1月唾液细胞外囊泡的miR-21-5p、miR-486-5p、miR-512-3p、miR-412-3p、miR-10b-5p的表达差异。结果OSCC患者的唾液细胞外囊泡中总蛋白浓度显著高于健康对照组,总RNA浓度在两组之间没有显著差异,而miR-21-5p和miR-10b-5p在OSCC患者中的相对表达量显著高于健康对照组;OSCC患者术前血液和唾液细胞外囊泡中miR-21-5p表达趋势基本一致,呈正相关;miR-21-5p在OSCC患者术后表达显著下调。结论唾液细胞外囊泡中miR-21-5p和miR-10b-5p可作为一种辅助OSCC诊断的标志物;而术后miR-21-5p的表达在手术效果评估中具有很好的参考价值。Objective To analyze the significance of salivary extracellular vesicle miRNA to diagnosis and postoperative evaluation of OSCC.Methods We compared the expressions of total miRNA of 17 OSCC patients and 11 healthy people,miR-21-5p of extracellular vesicles from blood and saliva of OSCC patients before operation,and miR-21-5p、miR-486-5 p、miR-512-3 p、miR-412-3 p、miR-10 b-5 p from salivary extracellular vesicles of OSCC patients before operation and one month after operation.Results(1)The total concentration of protein in salivary extracellular vesicles of OSCC patients were significantly higher than healthy controls,while that of RNA showed no significant difference,and the relative expression of miR-21-5p and miR-10 b-5 p were also significantly higher in OSCC patients.(2)The expression of miR-21-5p from blood correlated positively with that from saliva of OSCC patients before operation.(3)The expression of miR-21-5p of OSCC patients significantly decreased after operation.Conclusion MiR-21-5 p and miR-10 b-5 p from salivary extracellular vesicle can be considered as a supportive marker for diagnosis of OSCC,and the expression of miR-21-5p after operation provides great reference for postoperation evaluation.
