高效液相色谱法同时测定羚羊感冒片中7种活性成分的含量

摘　　要：目的建立高效液相色谱法(HPLC)同时测定羚羊感冒片中7种活性成分的含量。方法采用HPLC,同时测定羚羊感冒片中牛蒡苷、甘草苷、绿原酸、木犀草苷、连翘苷、连翘酯苷A和甘草酸铵的含量,采用Shim-pack GIST C_(18)色谱柱(250 mm×4.6 mm,5μm),流动相为乙腈(A)-0.05%磷酸溶液(B),梯度洗脱;检测波长:237 nm(检测甘草苷),245 nm(检测甘草酸铵),280 nm(检测连翘苷、牛蒡苷),328 nm(检测绿原酸、连翘酯苷A),350 nm(检测木犀草苷),流速1.0 ml/min,柱温:35℃,进样量:10μl。结果牛蒡苷、甘草苷、绿原酸、木犀草苷、连翘苷、连翘酯苷A、甘草酸铵进样量分别在0.350~7.000μg、0.136~2.720μg、0.204~4.080μg、0.220~4.400μg、0.074~1.480μg、0.090~1.800μg、0.096~1.920μg范围内与峰面积呈良好线性关系;平均加样回收率分别为99.2%,98.4%,99.2%,98.9%,99.1%,98.7%,98.7%,其RSD分别为0.35%,1.28%,0.65%,0.98%,1.12%,1.25%,1.09%。结论该方法准确、简单、有效,可用于同时测定羚羊感冒片中上述7种活性成分的含量。Objective To establish an HPLC method for the simultaneous determination of seven constituents in Lingyang Ganmao Tablets.Methods The determination was performed on Shim-pack GIST C_(18)column using acetonitrile-0.05%phosphoric acid at the flow rate of 1.0 ml/min as mobile phase of gradient elution.The detection wavelengths were set at 237 nm for liquiritin,245 nm for ammonium glycyrrhizinate,280 nm for arctiin and forsythin,328 nm for chlorogenic acid and forsythoside A,and 350 nm for cymaroside.The column temperature was set at 35℃and the injection volume was 10μl.Results The linear ranges of arctiin,liquiritin,chlorogenic acid,cymaroside,forsythin,forsythoside A and ammonium glycyrrhizinate were 0.350-7.000μg,0.136-2.720μg,0.204-4.080μg,0.220-4.400μg,0.074-1.480μg,0.090-1.800μg and 0.096-1.920μg,respectively.The recoveries of the seven constituents were 99.2%,98.4%,99.2%,98.9%,99.1%,98.7%and 98.7%,with RSDs of 0.35%,1.28%,0.65%,0.98%,1.12%,1.25%and 1.09%,respectively.Conclusion The method is accurate,simple and effective.It can be used for simultaneous determination of seven active constituents in Lingyang Ganmao Tablets.
