摘 要:目的了解矽肺大鼠的肝肾损伤及黄芩苷对矽肺大鼠的肝肾毒性作用。方法选用Wistar雄性大鼠50只,随机分为对照组、模型组和黄芩苷给药组,每组10只,采用一次性气管灌注SiO2的方法建立矽肺模型,连续经口灌胃黄芩苷(50、100、200 mg/kg)8周,检测各组大鼠肝肾脏器系数、血清中丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、碱性磷酸酶(ALP)、尿素氮(BUN)、肌酐(CRE)的含量及大鼠肝肾组织形态学变化。结果黄芩苷200 mg/kg组大鼠肾脏系数明显低于模型组,差异有统计学意义(均P<0.05)。黄芩苷200 mg/kg组大鼠血清中AST、ALP和BUN含量明显高于模型组,差异均有统计学意义(均P<0.05)。黄芩苷200 mg/kg组大鼠肾小球萎缩变小、肾小管明显萎缩且上皮细胞坏死。结论矽肺模型大鼠肝肾组织无明显损伤,一定剂量的黄芩苷对矽肺大鼠有明显的肝肾毒性作用。Objective To investigate the liver and kidney injury of silicosis rats and the effect of baicalin on liver and kidney toxicity in silicosis rats.Methods Totally 50 Wistar male rats were randomly divided into control group,model group and baicalin group,10 animals in each group.Silicosis model was established by one-time tracheal infusion of SiO2.Baicalin(50,100,200 mg/kg)was orally administered for 8 weeks.The rats in groups were tested for organ coefficient of liver and kidney,the serum levels of alanine aminotransferase(ALT),aspartate aminotransferase(AST),alkaline phosphatase(ALP),urea nitrogen(BUN),creatinine(CRE),and the liver and kidney tissue morphological changes.Results The renal coefficient of rats in 200 mg/kg baicalin group was significantly lower than that in model group,and the difference was statistically significant(P<0.05).The contents of AST,ALP and BUN in the serum of rats in 200 mg/kg baicalin group were significantly higher than those in model group,and the differences were statistically significant(all P<0.05).In 200 mg/kg baicalin group,the rats had glomerulus atrophy,renal tubule atrophy and epithelial cell necrosis.Conclusion There is no obvious damage to the liver and kidney tissues of silicosis rats.A certain dose of baicalin has obvious hepatorenal toxicity on silicosis rats.
