摘　　要：肿瘤的形成是由于细胞生长调控严重紊乱、染色体有丝分裂不稳定等多种因素共同导致细胞异常增生的结果。T细胞酪氨酸激酶(TTK)是纺锤体组装检查点(SAC)的核心部件,它能确保染色体向子细胞适当分布,平衡生长和分裂,是保证有丝分裂保真度和基因组稳定的一种蛋白激酶。当TTK过表达时,中心体增大,染色体不稳定,可导致肿瘤发生。TTK已被证实在胶质母细胞瘤、乳腺癌、肺癌、肝癌、结肠癌等多种恶性肿瘤中过表达。TTK在正常组织与癌组织中的差异性表达提示其具有临床诊断生物标记物的潜能,而TTK小分子抑制剂在动物模型中可以抑制肿瘤细胞的增殖,提示TTK具有肿瘤靶向治疗的潜力。本文将综述TTK在多种恶性肿瘤中的表达及临床意义的研究进展。Oncogenesis is the result of dysplasia of cells caused by a variety of factors,such as dysregulation of cell growth,unstable chromosome mitosis.T cell tyrosine kinase(TTK),a protein kinase,is the core component of spindle assembly checkpoint(SAC).TTK guarantees mitotic fidelity and genomic stability by distributing proper chromosomes to next generation cells and maintaining the balance between growth and division.Overexpressed TTK causes centrosome enlargement and chromosomal instability,and triggers tumorigenesis,such as glioblastoma,breast cancer,lung cancer,liver cancer,and colon cancer.The differential expression of TTK in cancer tissues and normal tissues suggests that its potential as a clinical diagnostic biomarker,while small molecule inhibitors of TTK can inhibit the proliferation of tumor cells in vivo,suggesting that TTK has potential for tumor-targeted therapy.This article summarized the research progress of TTK expression and clinical significance in a variety of human malignant tumors.
