摘 要:为了揭示设施连作不同果类菜根际土壤细菌群落结构和多样性变化,采用Illumina MiSeq测序平台进行细菌16S rRNA高通量测序分析,研究设施连作5 a的番茄、黄瓜、辣椒和茄子之间的根际土壤和露地土壤细菌群落结构和多样性差异,探讨设施连作的不同作物对土壤细菌群落结构和多样性的影响。结果表明:设施连作土壤中细菌多样性丰富,包括44个门、150个纲、364个目、575个科和1046个属,其中变形菌门、绿弯菌门、酸杆菌门、放线菌门、拟杆菌门和芽单胞菌门是土壤样品中细菌物种丰富度较高的6个门类,占所有细菌群落总数为露地84.56%、番茄83.02%、黄瓜82.21%、辣椒80.67%、茄子79.25%,4种作物根际土壤优势菌门丰富度占比均有显著差异并低于露地土壤;其中连作黄瓜变形菌门最高,放线菌门最低,分别占所有细菌群落总数31.53%,4.95%,根际土壤有益菌群高于其他果类菜和露地;连作番茄变形菌门丰度最高,酸杆菌门丰度最低,分别占所有细菌群落总数23.56%,5.16%,相对土壤质量最差。4种不同连作果类菜之间根系土壤细菌群落多样性均没有显著性差异,与露地相比连作黄瓜细菌群落多样性有显著性差异,表明不同果类菜根际土壤微生物多样性没有显著差异。为设施连作在内的许多农作物领域提供参考和可持续种植提供理论基础。In order to reveal the changes of bacterial community structure and diversity in rhizosphere soil of different fruit vegetables under greenhouse continuous cropping,the 16S rRNA gene of bacteria from the rhizosphere soil of planting tomato,cucumber,pepper and eggplant under continuous cropping for five years in a row and the open field as control,was sequenced by high throughput sequencing,and explore the effects of different crops in protected continuous cropping on soil bacterial community structure and diversity.The results showed that there were 44 phylum,150 classes,364 orders,575 families and 1046 genus of bacteria in greenhouse continuous cropping soil.The 6 phylum with high abundance were Proteobacteria,Chloroflexi,Acidobacteriota,Actinobacteriota,Bacteroidota and Gemmatimonadota,which accounted for 84.56%of the total bacterial community in open field,83.02%in tomato,82.21%in cucumber,80.67%in pepper and 79.25%in eggplant.The abundance of dominant bacteria in rhizosphere soil between four different crops showed a significant difference,and all of them were lower than open field.There were the highest Protrobacteria with 31.53%of the total bacterial communities and the lowest A ctionbacteriota with 4.95%after continuous cropping of cucumber,that showed the dominant bacterial population in rhizosphere soil of planting cucumber was higher than other vegetables and open field.While the soil quality of continuous cropping tomato,that the abundance of Proteobacteria was the highest and Acidobacteriota was the lowest,accounting for 23.56%and 5.16%of the total bacterial community,respectively.There was no significant difference in the bacterial community diversity of rhizosphere soil among the four continuous cropping vegetables,but there was significant difference between continuous cropping cucumber and open field.Which showed that the diversity of rhizosphere microorganisms in different crops was no significant difference.It would provide a theoretical basis for continuous cropping of different fruit vegetable
