摘 要:目的:研究红景天苷对癫痫持续状态大鼠海马神经细胞凋亡的影响。方法:50只SD大鼠随机分成5组,每组10只,分别为对照组、癫痫持续状态组(SE组,注射和红景天苷等量的DMSO)和A组、B组、C组(在造模前15 min分别腹腔注射0.5、1.0、1.5 g/kg红景天苷),腹腔内注射氯化锂和匹罗卡品构建癫痫模型,并测定大鼠癫痫发作潜伏期。取大鼠海马组织,采用相应试剂盒检测过氧化氢酶(CAT)、超氧化物歧化酶(SOD)、还原型谷胱甘肽(GSH)水平,TUNEL法检测细胞凋亡情况,Western blot法检测活化的Caspase-12(Cleaved-Caspase-12)、活化的Caspase-3(Cleaved-Caspase-3)、C/EBP同源蛋白(CHOP)、葡萄糖调节蛋白78(GRP78)蛋白表达水平。结果:与对照组比较,SE组大鼠海马组织中CAT、SOD、GSH水平均降低,神经细胞凋亡数升高,Cleaved-Caspase-12、Cleaved-Caspase-3、CHOP、GRP78蛋白表达水平升高(P<0.05)。与SE组比较,A组、B组、C组大鼠癫痫发作潜伏期延长,海马组织中CAT、SOD、GSH水平升高,神经细胞凋亡数减少,Cleaved-Caspase-12、Cleaved-Caspase-3、CHOP、GRP78蛋白表达水平降低(P<0.05)。结论:红景天苷可抑制癫痫持续状态大鼠海马神经细胞凋亡,作用机制可能与抑制氧化应激从而减少内质网应激有关。Aim:To investigate the effects of salidroside on hippocampal neuronal apoptosis in rats with status epilepsy.Methods:A total of 50 SD rats were allocated into five groups,the control group,the status epilepticus group(SE group,injecting the same amount of DMSO as salidroside),group A,group B,and group C(intraperitoneal injection of salidroside at 0.5,1.0,1.5 g/kg respectively 15 min before the establishment of the model).The models were made by intraperitoneal injection of lithium chloride and pilocarpine,and the latent period of epilepsy was evaluated.The levels of catalase(CAT),superoxide dismutase(SOD)and glutathione(GSH)were determined by the kits,TUNEL was used to detect apoptosis,and Western blot was used to detect Cleaved-Caspase-12,Cleaved-Caspase-3,C/EBP homologous protein(CHOP)and glucose regulatory protein 78(GRP78)proteins.Results:Compared with the control group,the levels of CAT,SOD,and GSH in the hippocampal nerve tissue of the SE group were decreased,the number of apoptotic cells increased,and the protein expression levels of Cleaved-Caspase-12,Cleaved-Caspase-3,CHOP,and GRP78 were increased(P<0.05).Compared with the SE group,the latent period of epilepsy in the group A,B,and C increased,the levels of CAT,SOD,and GSH increased,the number of apoptotic cells decreased,and the protein expression levels of Cleaved-Caspase-12,Cleaved-Caspase-3,CHOP,GRP78 were decreased(P<0.05).Conclusion:Salidroside could inhibit hippocampal neuronal apoptosis in rats with status epilepsy,and the mechanism may be related to inhibiting oxidative stress and reducing endoplasmic reticulum stress.
