摘 要:目的:研究沙棘总酚酸对胃癌细胞MGC-803增殖、迁移和细胞周期的影响机制。方法:采用CCK-8法检测沙棘总酚酸对MGC-803细胞增殖的影响,transwell和流式细胞术PI单染法检测沙棘总酚酸对MGC-803细胞迁移和细胞周期的影响,qRT-PCR技术检测Wnt/β-catenin信号通路中SFRP、Gsk-3β和E-cadherin的表达情况。结果:沙棘总酚酸对MGC-803细胞的增殖、迁移和细胞周期有明显抑制作用,呈浓度依赖性。qRT-PCR结果显示SFRP基因的表达上调和Gsk-3β、E-cadherin基因表达下调。结论:沙棘总酚酸抑制胃癌细胞的增殖、迁移,诱导细胞周期阻滞,作用可能与其调控Wnt/β-catenin信号通路中SFRP、Gsk-3β和E-cadherin基因表达量有关。Objective:To investigate the impact mechanism of total phenolic acids of Hippophae rhamnoides on the prolif-eration,migration and cell cycle of gastric cancer cell line MGC-803.Methods:CCK-8 assay was used to detect the effect of total phenolic acids of Hippophae rhamnoides on the proliferation of MGC 803 cells.Transwell and flow cytometry PI staining were used to detect the effect of total phenolic acid of seabbuckthorn on the migration and cell cycle of MGC-803 cells.qRT-PCR was used to detect the expression of SFRP,GSK-3βand E-cadherin in Wnt/β-catenin signaling pathway.Results:The total phenolic acids of Hippophae rhamnoides significantly inhibited the proliferation,migration and cell cycle of MGC-803 cells in a concen-trationdependent manner.qRT-PCR resuLts showed that the expression of SFRP gene was up-reguLated and the expression of GSK-3βand E-cadherin gene was down-reguLated.Conclusion:Total phenolic acid of Hippophae rhamnoides inhibits prolif-eration and migration of gastric cancer cells,and induces cell cycle arrest,which may be related to the reguLation of SFRP,GSK 3βand E-cadherin gene expressions in Wnt/β-catenin signaling pathway.
