摘 要:本研究旨在探讨饲粮添加香菇多糖对轮状病毒攻毒破坏断奶仔猪回肠屏障功能是否具有缓解作用。试验选取28头健康的21日龄断奶的“杜×长×大”仔猪,采用2×2因子试验设计,即香菇多糖处理(饲粮中不添加或添加84 mg/kg香菇多糖)和轮状病毒攻毒(灌服轮状病毒液或用于病毒培养的无菌培养液),轮状病毒攻毒于试验第15天早上进行。试验期为19 d。结果表明,轮状病毒攻毒提高了断奶仔猪的腹泻指数,显著提高了血清中丙二醛(MDA)含量及回肠黏膜中轮状病毒特异性抗体、白细胞介素-2(IL-2)和MDA含量(P<0.05),显著上调了回肠黏膜中B细胞淋巴瘤/白血病-2相关X蛋白(Bax)的mRNA相对表达量(P<0.05),显著降低了血清总抗氧化能力(T-AOC)及回肠黏膜中分泌型免疫球蛋白A、白细胞介素-4(IL-4)、干扰素-γ(IFN-γ)含量和T-AOC(P<0.05),显著降低了回肠黏膜中B细胞淋巴瘤/白血病-2(Bcl-2)、闭合蛋白(occludin)、闭锁小带蛋白-1(ZO-1)、黏液蛋白1(mucin 1)、黏液蛋白2(mucin 2)、猪β防御素1(pBD1)、猪β防御素2(pBD2)、猪β防御素3(pBD3)和Protegrins 1-5(PG1-5)的mRNA相对表达量以及回肠食糜中双歧杆菌、乳酸菌和总菌数量(P<0.05);而饲粮添加香菇多糖降低了轮状病毒攻毒仔猪的腹泻指数,并显著缓解了轮状病毒对断奶仔猪回肠黏膜中sIgA和细胞因子(如IL-2和IFN-γ)含量、肠道健康相关基因(如occludin、ZO-1、mucin 1、mucin 2、pBD2、pBD3和PG1-5)的表达及抗氧化能力的影响(P<0.05)。综上所述,饲粮添加香菇多糖可在一定程度上改善轮状病毒感染断奶仔猪的回肠屏障功能,这在香菇多糖缓解轮状病毒攻毒诱导腹泻中起到了重要的作用。This experiment was conducted to determine whether dietary lentinan can alleviate the ileal barrier function of weaned pigs challenged by rotavirus(RV).Twenty-eight healthy crossbred(Duroc×Landrace×Yorkshire)piglets weaned at 21 days of age were used in a 2×2 factorial design.The trial factors included diets(control diet or supplementing with 84 mg/kg lentinan diet)and RV challenge(oral gavage with RV or sterilized medium).On day 15,the RV challenge was carried out.The trial duration was 19 days.The results showed that RV infection increased the diarrhea index of weaned pigs,significantly elevated the serum malondialdehyde(MDA)content and the contents of rotavirus antibody,interleukin-2(IL-2)and MDA in ileal mucosa(P<0.05),significantly up-regulated the B-cell lymphoma/leukaemia 2-associated X protein(Bax)mRNA relative expression level in ileal mucosa(P<0.05),significantly decreased the serum total antioxidant capacity(T-AOC)and the contents of secreting immunoglobulin A(sIgA),interleukin-4(IL-4)and interferonγ(IFN-γ)as well as T-AOC in ileal mucosa(P<0.05),significantly down-regulated the mRNA relative expression levels of B-cell lymphoma/leukaemia 2(Bcl-2),zonula occludens 1(ZO-1),occludin,mucin 1,mucin 2,porcineβ-defensin 1(pBD1),porcineβ-defensin 2(pBD2),porcineβ-defensin 3(pBD3)and protegrins 1-5(PG1-5)in ileal mucosa(P<0.05),and significantly reduced the populations of Lactobacillus,Bifidobacterium and total bacteria in ileal digesta(P<0.05).Dietary lentinan reduced the diarrhea index of the RV-infected pigs,and significantly alleviated the effects of RV challenge on the contents of sIgA and cytokines(such as IL-2 and IFN-γ),the mRNA expressions of gut health related genes(including occludin,ZO-1,mucin 1,mucin 2,pBD2,pBD3 and PG1-5)and antioxidant capacity(P<0.05).These results suggest that dietary lentinan can improve ileum barrier function of weaned pigs infected with RV to some extent,which plays an important role in alleviating diarrhea induced by RV challenge.
