摘 要:目的:研究GPD2在非小细胞肺癌(non-small cell lung cancer,NSCLC)组织中的表达和临床意义,及GPD2基因对裸鼠成瘤能力的影响。方法:选取119例NSCLC肿瘤组织及配对正常组织,行GPD2免疫组化实验,并分析GPD2的表达与NSCLC患者临床病理特征的关系。使用RNA干扰慢病毒载体感染A549细胞,接种至裸鼠右侧腋窝,监测小鼠肿瘤生长情况,测量最终体积和重量。结果:肺癌患者肿瘤组织中GPD2的表达明显高于对照组(P<0.01)。III/IV期组织中GPD2表达明显高于I/II期(P<0.05),同时GPD2在淋巴结转移患者中高表达(P<0.05)。shGPD2细胞形成肿瘤体积明显小于shCtrl细胞形成肿瘤体积,且肿瘤生长速度远低于shCtrl细胞形成的肿瘤。结论:GPD2蛋白在NSCLC组织中表达显著高于癌旁组织,其表达水平与患者TNM分期、淋巴结转移均显著相关,可能参与了NSCLC的发生、发展。Objective:To study the expression and clinical significance of GPD2 in non-small cell lung cancer,and the effect of GPD2 gene on tumor formation in nude mice.Methods:To select 119 cases of non-small cell lung cancer tumor tissues and matched normal tissues,perform GPD2 immunohistochemistry experiments,and analyze the relationship between GPD2 expression and clinicopathological characteristics of NSCLC patients.Useing RNA interference lentiviral vector to infect A549 cells,inoculate them into the right axilla of nude mice,monitor the tumor growth of mice,and measure the final volume and weight.Results:The expression level of GPD2 in tumor tissues of lung cancer patients was significantly higher than that of the control group(P<0.01).The level of GPD2 in stage III/IV tissue was significantly higher than that in stage I/II(P<0.05),and GPD2 was highly expressed in patients with lymph node metastasis(P<0.05).The tumor volume formed by shGPD2 cells was significantly smaller than that formed by shCtrl cells,and the tumor growth rate was much lower than that formed by shCtrl cells.Conclusion:The expression of GPD2 protein in NSCLC tissue is significantly higher than that in adjacent tissues.Its expression level is significantly related to the TNM stage of NSCLC patients and lymph node metastasis,and may be involved in the occurrence and development of NSCLC.
