摘  要:目的探索ATP柠檬酸裂解酶(ATP citrate lyase,ACLY)在直肠癌组织中的表达情况及其临床意义,并通过基因沉默方法验证其细胞迁移功能。方法收集56例直肠癌患者病理组织,对直肠癌患者癌组织及癌旁组织标本进行ACLY免疫组化染色,并通过基因沉默方法及transwell实验在HCT116结直肠癌细胞株上干扰ACLY表达并验证对细胞迁移功能的影响。结果相比癌旁组织,直肠癌组织中ACLY表达显著增高,且肿瘤末期患者直肠癌组织ACLY表达逐渐加强。直肠癌组织中ACLY高表达的直肠癌病患者存期显著降低,差异有统计学意义(P<0.05)。沉默HCT116结直肠癌细胞中ACLY表达显著抑制结直肠癌细胞体外迁移,差异有统计学意义(P<0.05)。结论ACLY在结肠腺癌组织中高表达,与预后相关,干扰ACLY基因抑制结直肠癌HCT116细胞体外迁移,可能是直肠癌患者治疗的潜在靶点。Objective To investigate the expression of ATP citrate lyase(ACLY)in rectal cancer and its clinical significance,then evaluate its migration function by gene silencing in HCT116 cell line.Methods The expression of ACLY in 56 rectal cancer cases was investigated by immunohistochemical staining.The ACLY expression was interfered by gene silencing and its migration function was evaluated by the transwell method.Results Compared with the para-tumor tissues,the expression of ACLY was significantly higher in rectal cancer,and significantly increased in the rectal patients with advanced tumor stage,and the survival time of rectal cancer patients was significantly decreased in the high expression of ACLY patients,also had a statistical significance(P<0.05).Silence of ACLY expression in HCT116 colorectal cancer cell line can significantly inhibited the migration of rectal cancer cells in vitro,with the difference of statistically significant(P<0.05).Conclusion The high expression of ACLY are associated with the prognosis of rectal cancer patients,and the interference of ACLY gene can significantly inhibit the migration of HCT116 cells in vitro.It may be a potential target for the treatment of rectal cancer.
