摘  要:随着沙漠化面积的不断扩大,耐旱品种马铃薯的培育对生产越来越重要。本试验用PEG-6000模拟干旱,研究六个马铃薯组培苗生理指标的变化。结果表明,随PEG-6000浓度的升高,六个马铃薯品种的脯氨酸(Pro)含量、丙二醛(MDA)含量、过氧化物酶(SOD)活性和过氧化氢酶(CAT)活性都出现了先升高后降低的现象,且‘荷兰15’上升快,下降慢,说明其对干旱胁迫具有较强的耐受性。利用NCBI进行检索,得到马铃薯耐旱相关基因StAREB。生物信息学分析发现,StAREB总计编码453个氨基酸,多肽链属于非分泌型、疏水性蛋白。二级结构预测发现,该蛋白含有α-螺旋、延伸链和无规则卷曲分别为123、75和255处。本研究结果表明,六个品种中‘荷兰15’的耐旱能力最强,对选育耐旱马铃薯品种具有指导意义。With the continuous expansion of desertification area,the cultivation of drought-tolerant varieties of potato is in urgent need.In this study,we used PEG-6000 to simulate drought condition to study the physiological changes in six varieties of potato.As the concentration of PEG-6000 increased,we observed that the proline(Pro)content,malondialdehyde(MDA)content,peroxidase(SOD)activity and catalase(CAT)activity in six varieties increased first and then decreased later.Specifically,the variety'Holland15'increased most rapidly and decreased slowly,indicating that it had strong tolerance to drought stress.The drought tolerance gene StAREB was obtained by NCBI retrieval.Bioinformatics analysis showed that StAREB encoded 453 amino acids in total,and its polypeptide chain belonged to non-secretory and hydrophobic proteins.Secondary structure prediction showed that the protein contained 123α-helices,75 extended chains and 255 irregular curls,respectively.Thus,we concluded that'Holland15'is the most drought tolerance line among the six varieties,which could be used for drought-tolerant breeding in potato.
