摘 要:为了探究粗榧树皮和叶片中内生菌的群落组成与结构,本研究采用高通量测序方法对粗榧树皮和叶片中内生菌的群落结构与多样性进行分析。结果表明,粗榧树皮中细菌的ACE指数、Chao1指数和Shannon指数分别为169、170、3.58,均低于叶片,而真菌的ACE指数、Chao1指数和Shannon指数分别为281、283、4.19,均高于叶片,说明粗榧树皮的内生细菌群落丰富度及多样性比叶片低,内生真菌群落丰富度及多样性高于叶片。粗榧树皮和叶片中内生细菌在门水平上主要为变形菌门(Proteobacteria)、厚壁菌门(Firmicutes)、放线菌门(Actinobacteria)、蓝藻菌门(Cyanobacteria)和拟杆菌门(Bacteroidetes),其中变形菌门(Proteobacteria)为树皮和叶片中优势菌门;真菌门水平上主要为子囊菌门(Ascomycota)和担子菌门(Basidiomycota),子囊菌门(Ascomycota)为树皮和叶片中优势菌门。在属水平上,内生细菌主要为假单胞菌属(Pseudomonas)、乳球菌属(Lactococcus)、芽孢杆菌属(Bacillus)、寡养单胞菌属(Stenotrophomonas)、肠球菌属(Enterococcus)、不动杆菌属(Acinetobacter)等,假单胞菌属(Pseudomonas)均为树皮和叶片中的优势菌属;内生真菌主要有杯梗孢属(Cyphellophora)、帚枝霉属(Sarocladium)、德福里斯孢属(Devriesia)、Musicillium、Chaetothyriales_unclassified、Mycosphaerellaceae_unclassified、Sordariomycetes_unclassified、赤霉菌属(Gibberella)、Incertae_sedis_unclassified、链格孢属(Alternaria)、毛孢子菌属(Trichosporon)、Hannaella等,其中杯梗孢属(Cyphellophora)为粗榧树皮中的优势菌属,Mycosphaerellaceae_unclassified为粗榧叶片中的优势菌属。本研究结果可为进一步对海南粗榧内生菌的开发与利用提供理论依据。In order to explore the community composition and structure of endophytic bacteria and fungi in the bark and leaves of Cephalotaxus hainanensis,a high-throughput sequencing method was used in this study.The results showed that the ACE,Chao1 and Shannon indexes of bacteria in bark of C.hainanensis were 169,170 and 3.58 respectively,which were lower than those in leaves,while those of fungi were 281,283 and 4.19 respectively,which were higher compared with leaves.It showed that the abundance and diversity of endophytic bacteria in bark of C.hainanensis were lower than those in leaves,while those of endophytic fungi were higher.At the phylum level,the endophytic bacteria in bark and leaves of C.hainanensis were Proteobacteria,Firmicutes,Actinobacteria,Cyanobacteria and Bacteroidetes,and Proteobacteria was the dominant phylum;the endophytic fungi were Ascomycota and Basidiomycota,and Ascomycota was the dominant phylum in bark and leaves.At the genus level,endophytic bacteria were Pseudomonas,Lactococcus,Bacillus,Stenotrophomonas,Enterococcus and Acinetobacter,and Pseudomonas was the dominant genus in bark and leaves of C.hainanensis;the endophytic fungi were Cyphellophora,Sarocladium,Devriesia,Musicillium,Chaetothyrialesunclassified,Mycosphaerellaceaeunclassified,Sordariomycetesunclassified,Gibberella,Incertaesedisunclassified,Alternaria,Trichosporon and Hannaella,and Cyphellophora was the dominant genus in bark,while Mycosphaerellaceaeunclassified was the dominant genus in leaves.These results could provide theoretical bases for further development and utilization of the endophytes of C.hainanensis.
