摘  要:利用纤维素酶的水解作用和超声波的机械效应,协同辅助提取玉米黄色素,研究各提取条件对提取量的影响.通过单因素试验和正交试验优化玉米黄色素的提取条件,结果表明:影响提取效果的各因素的顺序由大到小依次是酶解温度、酶解时间、乙醇浓度、纤维素酶浓度;最佳提取条件为酶解温度60℃、酶解时间2.5 h、乙醇浓度85%、纤维素酶浓度1.4%、料液比1∶6、超声温度60℃、超声时间15 min时,玉米黄色素提取液的吸光度值可达0.552,提取效果最好.利用超声辅助酶法,不仅节省时间而且提高提取量,为玉米黄色素的提取研究提供科学基础.Maize yellow pigment was extracted by the hydrolysis of cellulase and the mechanical effect of ultrasonic wave.The effects of extraction conditions on the extraction rate were studied.The extraction conditions of corn yellow pigment were optimized by single factor experiment and orthogonal experiment.The results showed that the ultrasonic extraction rate of maize yellow pigment was better,and the primary and secondary factors affecting the extraction effect were as follows,enzymatic hydrolysis temperature,enzymatic hydrolysis time,ethanol concentration,cellulase concentration.According to the results of orthogonal test optimization,the optimal extraction condition was enzymatic hydrolysis temperature of 60℃,enzymatic hydrolysis time of 2.5 h,the concentration of ethanol 85%,the concentration of cellulase 1.4%,and ratio of material to liquid 1∶6(g/mL),the ultrasonic temperature of 60℃,and the ultrasonic time of 15 min,the absorbance value of corn yellow pigment was 0.552,and the extraction effect was the best.The use of assisted-ultrasound enzymatic hydrolysis not only saved time but also increased the extraction rate,to provide a scientific basis for the study of extracting Maize yellow pigment.
