猪源植物乳杆菌JL01调节巨噬细胞极化抗鼠伤寒沙门菌作用的研究

摘　　要：为探讨猪源植物乳杆菌JL01对巨噬细胞极化的作用,本试验使用猪肺泡巨噬细胞系3D4/21作为研究对象,采用JL01体外与3D4/21共培养,通过检测M1型与M2型巨噬细胞标志蛋白i NOS、Arg-1表达及细胞因子TNF-α、IL-6和IL-10的分泌情况,以明确JL01调节巨噬细胞极化的方向。通过免疫荧光及涂板计数,检测巨噬细胞对鼠伤寒沙门菌的吞噬杀伤效应。结果显示,JL01可以提高i NOS和Arg-1的表达,并促进TNF-α、IL-6和IL-10的分泌,但i NOS表达增强更为显著,且能增强巨噬细胞杀伤沙门菌的能力。上述结果表明,植物乳杆菌JL01能够促进巨噬细胞的M1型极化并增强对沙门菌的抵抗能力。In order to investigate the effect of Lactobacillus plantarum JL01 on macrophage polarization,3 D4/21 alveolar macrophage line was used as the research object in this experiment.JL01 was co-cultured with 3 D4/21 in vitro.The expression of i NOS and Arg-1 and the secretion of TNF-α,IL-6 and IL-10 in M1 and M2 macrophages were detected to determine the direction of JL01 regulating macrophage polarization.The phagocytosis and killing effect of macrophages on Salmonella typhimurium were detected by immunofluorescence and CFUS.The results showed that JL01 could increase the expression of iNOS and Arg-1,and promote the secretion of TNF-α,IL-6 and IL-10,especially,the expression of i NOS was increased more significantly than that of Arg-1,and the ability of macrophages to kill Salmonella was also enhanced.In conclusion,L.plantarum JL01 can promote the M1 polarization of macrophages to enhance the resistance to Salmonella.
