摘  要:目的:观察异氟醚暴露对癫痫慢性期成年小鼠齿状回不同发育阶段神经元的作用。方法:将匹鲁卡品诱导的癫痫模型小鼠分为对照组和异氟醚组,5-溴脱氧尿嘧啶核苷(Brd U)腹腔注射标记新生细胞,免疫荧光染色观察小鼠齿状回Brd U、Brd U+双皮质素(DCX)、Brd U+神经元核抗原(Neu N)和Brd U+同源异形盒基因转录因子-1(Prox-1)的表达,免疫组化染色观察小鼠齿状回Prox-1的表达及定位,显微镜下观察和采集图像,并进行细胞计数。结果:Brd U阳性细胞主要集中分布于颗粒下区(SGZ)和门区,颗粒细胞层也可见散在阳性细胞,而异氟醚组阳性细胞数明显少于对照组;Brd U+DCX免疫荧光双标阳性细胞主要分布于SGZ和颗粒细胞层,异氟醚组双标阳性细胞数与对照组比较无显著性差异;Brd U+Neu N、Brd U+Prox-1免疫荧光双标细胞主要分布于颗粒细胞层,双标细胞数在异氟醚组均显著少于对照组。Prox-1免疫组化染色结果显示阳性细胞主要分布于颗粒细胞层和门区,门区异位的颗粒细胞数在异氟醚组显著少于对照组。结论:异氟醚暴露可选择性地损伤癫痫慢性期小鼠齿状回SGZ来源的处于新生阶段的神经细胞、早期成熟神经元和成熟神经元。Objective: To observe the effect of isoflurane exposure on neurons at different developmental stages of dentate gyrus of adult mice with chronic epilepsy. Methods: The epilepsy model mice induced by pilocarpine were divided into the control group and the isoflurance group. The intraperitoneal injection of the 5-bromo-2’-deoxyuridine( Brd U)was adopt to label the cells born during the injection time point. Then the following slice staining was performed respectively: The immunostaining for Brd U-positive cells,the double-labeling immunofluorescence for Brd U + DCX,Brd U +Neu N and Brd U + Prox-1 as well as the immunohistochemical staining for Prox-1-positive cells. The pictures were captured and the positive cells were counted under microscope. Results: The Brd U-positive cells were mainly located in the SGZ and there were few positive cells in the hilus and granular cell layer. The number of Brd U-positive cells in isoflurance group was significantly less than the control group. Brd U + DCX double-labelled cells were located in the SGZ and granular cell layer. The number of Brd U + DCX double-labelled cells in isoflurance group was no statistical difference compared with the control group. Brd U + Neu N,Brd U + Prox-1 immunofluorescent positive cells were mainly distributed in the granular cell layer,and the number of Brd U + Neu N and Brd U + Prox-1 double-labelled cells in isoflurance group were significantly less than the control group,respectively. Prox-1 immunohistochemical staining showed that the positive cells were mainly distributed in the granular cell layer and hilus,and the number of ectopic granulosa cells in the hilus in isoflurane group was significantly less than that in the control group. Conclusion: Isoflurane exposure can selectively damage the nascent cells,early mature neurons and mature granular cells and reduce the heterotopic granular cells in the hilus of dentate gyrus with the chronic epilepsy mice.
