摘 要:目的通过人类癌症基因组图谱(TCGA)数据库筛选乳腺癌中关键的增强子RNA(eRNA),探讨其与乳腺癌患者预后的相关性。方法下载组织特异性增强子来源的长链非编码RNA(lncRNA)及其调控基因的列表,并下载TCGA数据库中相关的RNA测序数据和临床数据。通过生物信息学分析筛选与预后相关的eRNA,筛选标准为与肿瘤的总体存活率(Kaplan–Meier对数秩检验,P<0.05)和调控基因(相关系数r>0.4,P<0.001)具有显著相关性。结果与乳腺癌总体存活率相关性最强的eRNA是AP003059.2,基因共表达分析显示包括苹果酸酶3(ME3)在内13个基因的表达与AP003059.2显著相关,AP003059.2可能通过影响这些基因的表达,从而在乳腺癌发生中发挥作用。结论AP003059.2可能是乳腺癌中关键的eRNA,并且与乳腺癌的预后显著相关。Objective To identify the key enhancer RNA(eRNA)in breast cancer based on TCGA database and explore its correlation with the prognosis of breast cancer patients.Methods Tissue-specific enhancer-derived RNAs and their regulating genes previously predicted using the computational pipeline PreSTIGE,were considered as putative eRNA-target pairs.The relevant RNA sequencing and clinical data in the TCGA database were downloaded.The eRNAs related to prognosis were screened through bioinformatics analysis.The key eRNAs related to overall survival(Kaplan-Meier log-rank test,P<0.05)and the predicted target(correlation coefficient r>0.4,P<0.001)were identified.Results The analysis showed that the most relevant eRNA to the survival of patients was AP003059.2,and malic enzyme 3(ME3)and other 12 genes may be its regulated target.Conclusion Our results suggest that AP003059.2 is a key eRNA correlated with the prognosis of breast cancer patients.
