摘  要:目的探讨长链非编码RNA(lncRNA)RP11-169D4.1-001对喉鳞状细胞癌(LSCC)细胞增殖、凋亡、迁移等生物学行为的影响。方法使用慢病毒载体构建稳定过表达RP11-169D4.1-001的喉鳞癌细胞系,采用实时细胞分析、细胞克隆形成、流式细胞周期、流式细胞凋亡、细胞划痕实验、Transwell实验检测RP11-169D4.1-001在LSCC细胞增殖、凋亡、迁移中的作用。结果RP11-169D4.1-001在LSCC细胞增殖、迁移过程中发挥抑制作用。过表达RP11-169D4.1-001可抑制LSCC细胞增殖、迁移能力,将细胞周期阻滞在G0/G1期,诱导细胞凋亡。结论RP11-169D4.1-001在LSCC细胞增殖、凋亡、迁移等生物学行为中发挥抑癌作用,有望成为喉癌治疗的分子靶标。Objective To explore the effects of long non-coding RNA(lncRNA)RP11-169D4.1-001 on the proliferation,apoptosis,migration and other biological behaviors of laryngeal squamous cell carcinoma(LSCC)cells.Methods Lentiviral vector was used to construct a stable overexpressing laryngeal squamous cell carcinoma cell line with lncRNA RP11-169D4.1-001.Real-time Cell Analyzer(RTCA),cell cloning,flow cytometry,scratch assay and Transwell migration assay were used to detect the effects of RP11-169D4.1-001 on the growth,proliferation,apoptosis and migration of laryngeal squamous cell carcinoma,respectively.Results lncRNA RP11-169D4.1-001 inhibited the proliferation and migration of laryngeal squamous cell carcinoma cells.The overexpression of RP11-169D4.1-001 inhibited the proliferation and migration of ASCC cells,arrested the cell cycle in G0/G1 phase,and induced cell apoptosis.Conclusion lncRNA RP11-169D4.1-001 may play an anti-cancer role in proliferation,apoptosis,migration and other biological behaviors of LSCC cells,and is expected to be a molecular target for the treatment of laryngeal cancer.
