摘　　要：目的探讨miR-141-3p对胃癌(GC)细胞迁移与侵袭的影响,并揭示其潜在的分子机制。方法用RT-qPCR分别检测临床组织样本、正常胃黏膜上皮细胞系GES-1和胃癌细胞系AGS中miR-141-3p的表达。在AGS细胞中转染miR-141-3p mimic后,用Transwell小室法检测细胞的迁移和侵袭;荧光素酶报告基因实验检测miR-141-3p是否通过靶向程序性细胞死亡蛋白1(PDCD1)发挥其生物学作用;慢病毒-PDCD1感染已过表达miR-141-3p的AGS细胞,随后检测细胞的迁移和侵袭。结果与癌旁组织和GES-1细胞比较,GC组织和AGS细胞中miR-141-3p的表达明显降低(P<0.01);过表达miR-141-3p可显著抑制AGS细胞的迁移和侵袭(P<0.01);miR-141-3p可通过直接结合PDCD1 mRNA的3′-UTR,从而负调节PDCD1的表达;过表达PDCD1能逆转过表达miR-141-3p对AGS细胞的迁移和侵袭的抑制作用(P<0.01)。结论miR-141-3p通过下调靶基因PDCD1来抑制GC细胞的迁移和侵袭。Objective To investigate the role of miR-141-3p on the migration and invasion of gastric cancer(GC)cells,and to reveal underlying molecular mechanism.Methods The expression of miR-141-3p in clinical tissue samples,normal gastric mucosal epithelial cell line(GES-1)and gastric cancer cell line(AGS)was detected by RT-qPCR.miR-141-3p mimic was transfected into AGS cells,and then the migration and invasion of cells were detected by Transwell assay.The luciferase reporter experiment was applied to detect potential biological effects of miR-141-3p by targeting at programmed cell death protein 1(PDCD1).AGS cells that had over-expressed miR-141-3p were then infected with lentivirus PDCD1,and cell migration and invasion were subsequently detected.Results Compared with adjacent tissues and GES-1 cells,the expression of miR-141-3p in gastric cancer tissues and AGS cells was significantly decreased.Over-expression of miR-141-3p significantly inhibited migration and invasion of AGS cells.miR-141-3p negatively regulated PDCD1 expression by directly binding to the 3′-UTR of PDCD1 mRNA.Over-expression of PDCD1 reversed inhibitory effect of miR-141-3p over-expression on the migration and invasion of GAS cells.Conclusions miR-141-3p inhibits the migration and invasion of GC cells by down-regulating the target gene PDCD1.
