摘　　要：目的建立地榆方中没食子酸的含量测定方法,优选地榆方中地榆的提取工艺。方法采用高效液相色谱法对地榆方中没食子酸进行测定;以液料比、提取溶剂、提取时间为考察因素,以没食子酸含量为评价指标,采用L9(34)正交试验设计,优选地榆的提取工艺。结果没食子酸在101.3~607.79 ng(R^(2)=0.9999)范围内与吸收度呈良好的线性关系,平均加样回收率为91%,RSD为0.86 %。地榆的最佳提取工艺为:液料比1:100,10 %盐酸作为提取溶剂,回流提取3h,出膏率约为39.5 %。结论含量测定方法和优选的工艺稳定可行,可用于地榆方的质量控制和提取。Objective To establish a method for determining the content of gallic acid in Diyu Prescription,and optimize the extraction process of Radix Sanguisorba.Methods The content of gallic acid in Diyu Prescription was determined by high performance liquid chromatography(HPLC).The extraction process was optimized by orthogonal experimental design of L9(34),with the ratio of liquid to solid extraction solvent and extraction time as the factors,and gallic acid content as the evaluation index.Results There was a good linear relationship between gallic acid and absorbance in the range of 101.3~607.79 ng(R^(2)=0.9999);the average recovery was 91% and RSD 0.86%;the optimal extraction process of radix sanguisorba was as follows:the liquid-solid ratio was 1:100,10% hydrochloric acid was used as the extraction solvent,the reflux extraction lasted for 3 h,and the paste yield was about 39.5%.Conclusion The optimized process and method are simple and feasible,and can be used for the quality control and extraction of Diyu Prescription.
