摘  要:为研究茶多酚(TP)对H_(2)O_(2)诱导U251细胞氧化损伤的保护作用,试验分为正常组、氧化损伤模型组、TP低(25 mg/mL)剂量组、中(50 mg/mL)剂量组、高(100 mg/mL)剂量组,分别通过MTT法、Hoechst 33342染色法、流式细胞分析法和ELISA法测定TP对H_(2)O_(2)诱导下U251细胞存活率、细胞形态、凋亡率和胞内活性氧(ROS)水平、丙二醛(MDA)和胞内抗氧化酶活性的影响。结果表明:选择H_(2)O_(2)浓度和处理时间分别为400μmol/L和24 h来构建U251细胞氧化损伤模型;TP浓度为0~100 mg/mL对U251细胞无毒性作用;中、高剂量组TP能显著抑制H_(2)O_(2)造成的U251细胞存活率降低(P<0.05);低、中、高剂量组TP能显著降低细胞凋亡率、MDA、胞内ROS水平和荧光强度(P<0.05);同时中、高剂量组TP能显著提高胞内总抗氧化能力(T-AOC)及谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)和过氧化氢酶(CAT)活性。TP对H_(2)O_(2)诱导U251细胞氧化损伤具有保护作用,其作用可能与增强细胞内抗氧化酶活性,减少脂质发生过氧化,清除细胞内过量的ROS及抑制细胞凋亡有关。The aim of this study was to investigate the tea polyphenols(TP)on hydrogen peroxide(H_(2)O_(2))-induced oxidative damage in U251 cells.The experiment was divided into normal group,oxidative damage model group,low(25 mg/mL),medium(50 mg/mL)and high(100 mg/mL)dose groups of TP.The MTT,Hoechst 33342 staining,flow cytometry and ELISA were employed to determine the effects of TP on the survival rate,cell morphology,apoptosis rate,ROS levels,malondialdehyde(MDA)and intracellular antioxidant enzyme activities of U251 cells induced by H_(2)O_(2),respectively.The results showed that H_(2)O_(2)concentration and treatment time were 400μmol/L and 24 h respectively to establish U251 cells oxidative damage model.When the concentrations of TP were in the range of 0~100 mg/mL,it had no toxic effect on U251 cells.TP in middle and high dose groups significantly inhibited the decrease of U251 cell survival rate caused by H_(2)O_(2)(P<0.05);TP in low,middle and high dose groups substantially reduced the apoptosis rate,MDA,ROS levels and fluorescence intensity(P<0.05);At the same time,TP in the middle and high dose groups could prominently improve the total antioxidant capacity(T-AOC),glutathione peroxidase(GSH-Px),superoxide dismutase(SOD)and catalase(CAT)activities.TP could protect U251 cells from oxidative damage induced by H_(2)O_(2),which may be related to enhancing the activity of antioxidant enzymes,reducing lipid peroxidation,eliminating excessive ROS and inhibiting apoptosis.
